Embodiment 1

[0022] To investigate the effect of the content of marine Rhodotorula extract on the cultivation of Alexandrium microminis.

[0023] The algae liquid of Alexandrium microbiota in the logarithmic growth phase was inoculated in a 20L open raceway pool reactor, and the culture medium included each component of the following mass concentration: NaNO 3 1.6g / L, K 2 HPO 4 ·3H 2 O is 0.03g / L, MgSO 4 ·7H 2 O is 0.06g / L, ZnSO 4 0.03g / L, CoCl 2 0.002g / L, MnCl 2 0.04g / L, FeCl 3 ·6H 2 O is 0.004g / L, Na 2 EDTA·2H 2 O is 0.002g / L, citric acid is 0.005g / L, natural seawater; the initial density of Alexandrium microbiota after inoculation is 1×10 6 Individual / mL, adding different contents of marine rhodotorula extract for culture experiments; experiments were carried out at 30°C, and the light intensity was 350μmol / m 2 / s, the light-dark cycle 24h, the light-dark time ratio is 14:10 (hours), after culturing for 6 days, observe and count the quantity of microscopic Alexandrium and th...

Embodiment 2

[0029] Investigate the effects of temperature and light intensity on the cultivation of Alexandrium minutica.

[0030] The algae liquid of Alexandrium microbiota in the logarithmic growth phase was inoculated in a 20L open raceway pool reactor, and the culture medium included each component of the following mass concentration: NaNO 3 1.6g / L, K 2 HPO 4 ·3H 2 O is 0.03g / L, MgSO 4 ·7H 2 O is 0.06g / L, ZnSO 4 0.03g / L, CoCl 2 0.002g / L, MnCl 2 0.04g / L, FeCl 3 ·6H 2 O is 0.004g / L, Na 2 EDTA·2H 2 O is 0.002g / L, citric acid is 0.005g / L, natural seawater; the initial density of Alexandrium microbiota after inoculation is 1×10 6 Individual / mL, add 80mg / L marine rhodotorula extract to carry out culture experiments; culture light-dark cycle 24h, light-dark time ratio is 14:10 (hours), culture 6 days later, use hemocytometer method to count microscopic Alexander The number of algae and the number of miscellaneous bacteria and pests are observed and counted, and the results are sh...

Abstract

The invention discloses a culture method for alexandtium minutum capable of generating gonyautoxin. Conventional culture method and conditions are adopted for culturing alexandtium minutum, and a marine red yeast extract is added into a culture medium in the initial culture stage of alexandtium minutum. A preparation method for the marine red yeast extract comprises the following steps: taking drymarine red yeast powder, and then adding distilled water, (S)-(+)-3-(1-naphthoxy)-1-phenyl-1-propyl alcohol and citric acid, preparing into a suspension and ultrasonically treating; properly regulating pH and temperature of the solution, adding papain and performing enzymolysis; centrifuging, taking supernate and freeze-drying, thereby acquiring the marine red yeast extract. The invention has thebeneficial effects as follows: the invention provides a simple and efficient culture method for alexandtium minutum capable of generating gonyautoxin, the culture process is unlikely to result in pollution, and the alexandtium minutum is high in growth rate and high in toxin yield.

Description

technical field [0001] The invention relates to the field of biotechnology, in particular to a method for cultivating Alexandrium microbiota producing gonylin. technical background [0002] Since the first discovery of Saxitoxin STX in 1957, a total of 57 paralytic shellfish toxins (PSTs) homologues have been recorded. [0003] Paralytic shellfish toxins are a class of alkaloids with nerve paralyzing effects. The poisoning mechanism is that the toxin binds to the structure of sodium ion channels, hindering the flow of sodium ions. There is no antidote after poisoning, and the only rescue method is artificial respiration and infusion. Because of their strong toxicity, colorless, tasteless and paralyzing effects, PSTs have the possibility of being used in biochemical weapons in the military, and as precursors for exploring new anesthetics in medical treatment. Some PSTs toxin standards are sold, but the price is expensive. [0004] Paralytic shellfish toxin has a wide range...
