Embodiment 1

[0015] Detect the breeding eggs produced by female ducks containing the GFP fluorescent gene on the W chromosome, using the device of the present invention, the specific implementation steps and results are as follows;

[0016] 1. Collect 10 breeding eggs produced by mating female ducks containing the GFP fluorescent gene on the W chromosome and male ducks without GFP, and remove the stains on the surface of the eggshells for inspection.

[0017] 2. The egg to be inspected should be fixed horizontally, and should be still for a few seconds, so that the inside of the blastodisk is completely static, and the blastodisc can be freely rotated on the top.

[0018] 3. Select 20 positions on the eggshell on a plane perpendicular to the longitudinal axis of the egg, and perform 509nm fluorescence measurement. The correlation between the position of the fluorescence measurement and the fluorescence intensity has two unique forms. In the first form, no peaks appear; in the second form, ...

[0021] Detect the breeding eggs produced by female ducks containing the GFP fluorescent gene on the Z chromosome, using the device of the present invention, the specific implementation steps and results are as follows;

[0022] 1. Collect 10 breeding eggs produced by the mating of female ducks containing the GFP fluorescent gene on the Z chromosome and male ducks without GFP, and remove the stains on the surface of the eggshells for inspection.

Abstract

The invention discloses a photoelectric detection method for poultry gender. The photoelectric detection method comprises the following steps: horizontally fixing a to-be-detected egg collected by mating of a female duck with W chromosome containing GFP or a female duck with Z chromosome containing GFP and a male duck without GFP, carrying out standing for a few seconds until the interior of a blastoderm is completely static, and allowing the blastoderm to be able to freely rotate at the top part; and selecting 20 to 50 sites on an eggshell in a longitudinal axis plane perpendicular to the egg, and carrying out fluorescence measurement, wherein a correlation between a fluorescence measurement position and fluorescence intensity comprises two unique forms. The photoelectric detection methodprovided by the invention has the following beneficial effects: identification of a female embryo egg can be carried out under the condition of no damage to a breeding egg, is fast and effective andhas an accuracy rate reaching 100%.

Description

technical field [0001] The invention belongs to the technical field of biology and relates to a photoelectric detection method for poultry sex. Background technique [0002] Female selection of avian organisms is of great value in improving production efficiency in the poultry industry. The production performance of meat and layer birds is high. The large difference in metabolism of these two means that male layer birds are not productive. Now, males are screened after birth and immediately euthanized. This has great moral problems and economic losses for the entire industry. [0003] In our experimental animals for gender identification, the marker fluorescent gene has been positioned to a safe position on the female Z chromosome (Z*) by technical means in advance, and Z*W individuals are obtained, and then crossed with ZZ (male) to produce the following offspring : ZW (female), Z*Z (male), ZZ* (male), ZW (female). In this case, the marker gene on the female Z chromosome...
