Problems solved by technology

[0004] In summary, the problems in the prior art are: 1. The mycelium of black fungus has a certain tolerance to high temperature, and the mycelium can normally resume growth after being affected by a short-term high temperature
In the current mycelium high temperature resistance screening method, the heat treatment culture time is short, the high temperature condition is single, and the adaptability and stability of mycelium from low temperature to high temperature are lacking, so the high temperature resistant strain

Abstract

The invention belongs to the technical field of strain screening, and discloses an indoor screening method of high-temperature-resistant auricularia auricula-judae strains. The method comprises the steps of culturing the strains for 7 days under the condition of six temperature gradients, wherein each treatment is repeatedly carried out for three times; inoculating strain mycelial discs, which areactivated for 5 days, into the center of a PDA integrated medium, culturing for different periods of time at the temperature of 40 DEG C, then taking out and recovering normal culture; after the tested strains are activated for 5 days, selecting the mycelial discs at the edge of a bacterial colony, inoculating the selected mycelial discs into the center of a PDA integrated plate medium for culturing, and exercising at high temperature; after the tested strains are activated and cultured for 8 days, preparing the strains into cultivation strains; after bags are fully filled with the cultivation strains, inoculating the cultivation strains into a fruiting bag culture medium for culturing for about 50 days; respectively putting the fruiting bags in an environment with suitable fruiting conditions for producing auricularia auricula-judae. After the method is adopted, the strains which contain mycelia with better high temperature resistance and can normally bud and produce the auriculariaauricula-judae can be screened out, and a germplasm resource material with better high temperature resistance can be obtained; therefore, a foundation is laid for breeding the good high-temperature-resistant auricularia auricula-judae variety suitable for being cultivated in Guangxi region in the future.

Description

technical field [0001] The invention belongs to the technical field of strain screening, and in particular relates to an indoor screening method for high-temperature-resistant cloud ear strains. Background technique [0002] The fungus cultivated in Guangxi has a history of more than 500 years and is one of the four major edible fungi in Guangxi. Due to the stable market price of black fungus in recent years and obvious economic benefits, its cultivation area is expanding day by day. At present, most of the black fungus varieties mainly planted in Guangxi come from the medium and low temperature varieties in the north or Zhejiang and Fujian. Medium and low temperature varieties are cultivated in Guangxi. In warm winter weather, rotten bags are prone to occur, tides decrease, and yield and quality decline. Cloud fungus is a unique black fungus variety in Guangxi. The ear piece is brown, shiny, translucent, tender and smooth in the mouth, and has an excellent taste. Because ...
