Embodiment 1

[0030] 1) Extraction of the total genome: Use phenol and chloroform extraction methods to take about 100mg of muscle tissue samples, cut them into pieces and place them in a sterilized 2.0ml EP tube;

[0031] 2) Add 1ml of DNA extraction solution and 4uL of 20mg / ml RNaseA to the EP tube, shake well and place in a water bath at 37°C for 1 hour;

[0032] 3) Take out the EP tube, add 5uL 20mg / ml proteinase K, shake well, and place in a water bath at 50°C for about 2-3 hours until the muscle is completely digested, during which time shake up and down every 10 minutes;

[0033] 4) Take out the EP tube, after cooling to room temperature, add an equal volume of saturated phenol, and gently shake up and down for about 7 minutes until the water phase and the phenol phase are mixed into an emulsion;

[0034] 5) Centrifuge at 10000rPm for 15min at 10°C, take out the upper aqueous phase and transfer it to another 20ml EP tube;

[0035] 6) Repeat the phenol extraction once or twice;

Abstract

The invention discloses a primer and method for amplifying a magnetic induction receptor protein gene ISCA1 fragment of a grey-capped woodpecker. The primer comprises a first primer pair and a nested PCR primer, and the first primer pair comprises PIC-2L-F with the sequence number of SEQ ID NO.1 and PIC-2L-R with the sequence number of SEQ ID NO.7. The method comprises the following steps: (1) extracting the total genome DNA of a sample to be detected; (2) performing PCR amplification on the total genome DNA by the first primer pair; (3) performing secondary amplification on the product after amplification in the step (2) by the nested PCR primer; and (4) detecting the product after secondary amplification in the step (3). The primer and method can accurately, simply and quickly acquire a long fragment sequence of the ISCA1 gene of a grey-capped woodpecker.

Description

technical field [0001] The invention belongs to the technical field of biological detection, and in particular relates to a primer and a method for amplifying the ISCA1 fragment of the star-headed woodpecker magnetoreceptor protein gene. Background technique [0002] Many organisms use the geomagnetic field to navigate or migrate, such as the migration of migratory birds and the transmission of information by carrier pigeons. As early as 1978, the "free radical pair theory" model proposed by SchuLten et al. believed that the magnetoreceptor is likely to come from a blue light receptor protein called Cryptochrome (Cry), and this process involves electrons under a magnetic field quantum chemical reaction, and requires the participation of the visual system. This model later became the prototype of many theoretical works, gradually improved by Ritz and Wiltschkos et al., and Cry protein has been the only magnetoreceptor protein candidate for decades. Until June 2015, Vidal-Ga...
