Problems solved by technology

Although interleukin-17A plays a role in expanding the immune defense response to protect the body in the process of host anti-infection and tissue repair, interleukin-17A is highly expressed in many autoimmune disease patients and tumor patients, because it can Induces the expression of many inflammatory factors, and excessively high interleukin-17A levels play a role in worsening the pathological development of the disease

Embodiment 1

[0038] Example 1, Discovery of the relationship between interleukin 17 and influenza virus

[0039] 1. Cell culture and passage

[0040] 293T and A549 cells were cultured in DMEM containing 10% FBS and 1‰ double antibody at 37°C and 5% CO 2 in the incubator.

[0041] 1. When the cells are cultured to a confluence of more than 95%, they are ready to be subcultured;

[0042] 2. Discard the original medium, wash the residual medium and a small amount of dead cells on the cell surface with PBS;

[0043] 3. Add an appropriate amount of trypsin, and digest the corresponding time according to the cell adhesion ability. Observe under an optical microscope, and discard the trypsin after the cells become round;

[0044] 4. Add fresh medium, blow the cells gently into a cell suspension, pipette an appropriate amount of cell suspension into a new cell culture dish, add fresh medium to a total volume of 8-10mL, and store at 37°C, 5% CO 2 The incubator is placed for cultivation;

Embodiment 2

[0097] Embodiment 2, application of interleukin-17 in inhibiting the growth of influenza virus

[0098] The amino acid sequence of IL-17A precursor is sequence 1, and the amino acid sequence of mature peptide is sequence 2.

[0099] 1. Incubation of A549 cells with IL17A

[0100] A549 cells were divided into the following two groups for culture:

[0101] A549 cells to be infected: A549 cells (recorded in "S-S Wang, Zh-D Zhao, Y-H Bi, L Sun, X-LLiu*, W-J Liu*.2013.Tyrosine 132 phosphorylation of influenza A virus M1protein is crucial for virus replication by controlling its nuclear import.Journal of Virology, 87 (11): 6182-91 ", the public can obtain from the Institute of Microbiology, Chinese Academy of Sciences.) Cultivate in DMEM cell culture fluid for 24h, to 90% confluence;

[0102] Incubation of A549 cells to be infected with different concentrations of IL17A: A549 cells were cultured in a cell culture medium containing IL17A for 24 hours until 90% confluence.

Abstract

The invention discloses application of interleukin 17 in resisting of infection of influenza virus. The invention provides application of IL-17 protein in preparation of products for inhibiting influenza virus from infecting cells, application of IL-17 protein in preparation of influenza virus resisting products, and application of IL-17 protein in application of products for inhibiting influenza virus from growing in cells. The experiment shows that interleukin 17 can be used for resisting influenza virus from infecting the cells and has a large value on preventing and control of influenza virus.

Description

technical field [0001] The invention relates to the field of biotechnology, in particular to the application of interleukin 17 in resisting influenza virus infection. Background technique [0002] There are many types of interleukins with different functions, but a new type of interleukin discovered by recent studies plays an important role in the occurrence and development of inflammatory reactions. This type of interleukin is named interleukin-17. Some controversial inflammatory phenomena have provided reasonable explanations, and at the same time, people have a more detailed understanding of inflammatory signaling networks. Scientists first discovered interleukin-17A from mouse T-cell lymphoma cells. Through bioinformatics methods, they soon found that there were five other genes homologous to interleukin-17A in the genome. Therefore, people identified these six The two homologous cytokines were classified into a new interleukin family: the interleukin-17 family. The cy...
