Abstract

The invention relates to an external rooting culture method of endangered relict tree species yew seed tissue culture seedlings. The method comprises the following steps: disinfecting surfaces of yew seeds, inoculating the disinfected yew seeds to an MS medium to make 50% of the seeds germinate 30d later, taking terminal buds of aseptic seedlings with hypocotyl with the length of 1.5cm as explants, inducing cluster buds in a medium I to make the induction rate reach 80%, cutting off new terminal buds and stem segments with lateral bud, subculturing in a medium II to obtain subcultured buds with the subculture multiplication coefficient of 7.5, repeatedly cutting off subcultured bud terminal buds and bud segments, and carrying out propagation and propagation expanding in the medium II to obtain a large amount of propagated subcultured buds; separately cutting off 4-6cm length terminal buds, carrying out rapid dipping treatment in a 500mg.L<-1> alpha-naphthylacetic acid solution, transplanting the obtained buds to an aperture disk, carrying out external rooting, and covering with a plastic aperture disk cover to carry out moisture retention; and allowing the rooting treatment to be carried out for 60d to make the rooting rate reach 64%, and transplanting rooted seedlings to a nursery, wherein the transplanting survival rate reaches 95%.
