Embodiment

[1306] Treatment of patients with recurrent malignant glioma or progressive secondary brain tumors with dianhydrogalactitol

[1307] Brain tumors are among the most therapeutically challenging malignancies. The median survival of patients with recurrent disease of glioblastoma multiforme (GBM) is <6 months. Tumors that have spread to the brain, central nervous system (CNS) metastases have developed into a major contributor to cancer mortality based on the inability of improved systemic therapy to reach tumors.

[1308] The first-line systemic therapy is temozolomide but due to O 6- Resistance to methylguanine-DNA methyltransferase (MGMT) activity has been implicated in adverse outcomes. Such resistance greatly reduces survival rates.

[1309] Dianhydrogalactitol is a novel bifunctional N that easily crosses the blood-brain barrier and accumulates in brain tissue 7 DNA-alkylating agent. Dianhydrogalactitol causes cross-linking of DNA strands at N 7 - Guanine (E. Institóri...

Abstract

Methods and compositions suitable for the treatment of malignancies such as recurrent glioma and progressive secondary brain tumor are disclosed. These methods employ a hexitol derivative such as dianhydrogalactitol, a derivative or analog of dianhydrogalactitol, diacetyldianhydrogalactitol, or a derivative or analog of diacetyldianhydrogalactitol. The compositions can include such hexitol derivatives.

Description

[0001] [Citation to Related Application] [0002] This application asserts U.S. Provisional Patent Application Serial No. 61 / 829,739 filed on May 31, 2013 by K.C.Shi et al. Uses for gliomas or progressive secondary brain tumors". This application is incorporated herein by reference in its entirety. technical field [0003] The present invention is directed to methods and compositions comprising dianhydrogalactitol or its analogs or derivatives for treating malignant glioma or progressive secondary brain tumor. Background technique [0004] For many of the life-threatening diseases that afflict mankind, the research and identification of cures are still a process of experience and sometimes chance. Although there have been many advances from basic scientific research to improvements in actual patient management, there is still a great setback in the rational and successful discovery of effective therapies, especially for life-threatening diseases such as cancer, inflammatio...
