Embodiment Construction

[0015] 1. The inventor cloned the coding region of the maize ZmGolS2 gene by PCR method, and constructed the plant expression vector of the gene.

[0016] Dehydration treatment was performed on hydroponic three-leaf stage corn seedlings. RNA was extracted from leaves dehydrated for 2 hours and reverse transcribed into cDNA.

[0017] Using cDNA as a template, the coding region of ZmGOLS2 was amplified by PCR with the upstream primer 5'-CGGGATCCTGGCTCCCGAGCTGATGACAGCCAAG-3' and the downstream primer 5'-GCTCTAGACAAACACTCTAGCCCAATTCCTGACC-3'.

[0018] The amplification program was: pre-denaturation at 94°C for 5 min; denaturation at 94°C for 30 s, annealing at 55°C for 30 s, extension at 72°C for 1 min and 20 s, 35 cycles; final extension at 72°C for 8 min.

[0019] The amplified product was gel-cut and recovered, and the fragment was digested with BamHI and XbaI, and then combined with the pCsGFP vector (for stable transformation of Arabidopsis, see figure 1 )connect. The GFP ...

Abstract

The invention relates to the application of a corn ZmGolS2 gene in improving the drought resistance, high temperature resistance and salt resistance capability of plants. The inventor develops a method of increasing the content of galactinol and raffinose in plant leaves by virtue of overexpression of a corn GRMZM5G872256 gene (named as ZmGoLS2) and then improving the drought resistance, high temperature resistance and salt resistance of the plants. The corn ZmGolS2 gene is cloned by use of a PCR method by virtue of homological cloning. An expression vector for regulating and controlling the gene by use of a 35S promoter is constructed. The vector is transferred into the plant Arabidopsis by use of an agrobacterium tumefaciens mediated method, and then the content of galactinol and raffinose in the transgenic plant leaves can be remarkably increased, and the drought resistance, high temperature resistance and salt resistance capability of the transgenic plants can be remarkably improved. The gene-overexpressed plants grow normally under normal conditions and are superior to the known stress-resistant gene ZmDREB2A overexpressed plants in performance. The growth of the gene is superior to the growth of the control under the stress conditions of drought, high temperatures and high salinity, and is similar to the growth of ZmDREB2A overexpressed plants.

Description

technical field [0001] The invention relates to the application of the corn ZmGolS2 gene, in particular to the application of the corn ZmGolS2 gene to improve the ability of plants to resist drought, high temperature and salt. Background technique [0002] There are many reports on improving plant drought resistance, heat resistance, and salt resistance by regulating the expression of related genes. However, most transgenic plants are inhibited in plant growth under normal conditions, so so far, no related genes have been used in commercial production. There are very few reports on improving plant stress resistance by regulating raffinose metabolism genes. Himuro reported in 2014 that overexpression of the Arabidopsis AtGolS2 gene in Brachypodium distachyon enhanced the drought resistance of transgenic plants (1). Zhuo reported in 2013 that overexpression of the Medicago sativa MfGolS1 gene in tobacco increased the content of galactinitol and raffinose in transgenic tobacco...
