specific Embodiment

[0090] The present invention introduces the DNA high-throughput sequencing method into the technology for measuring the proportion of fetal free DNA in pregnant women's free DNA, and the flow of the specific application process is further described in conjunction with specific examples.

[0091] 1. Materials

[0092] The samples provided were 4 cases of peripheral blood of pregnant women with unknown fetal cell-free DNA concentration, including 2 cases of male fetuses and 2 cases of female fetuses. DNA concentrations were compared.

[0093] High-throughput sequencing reagents are mainly composed of 3 kits, library construction kit (amplification library construction kit and ligation library construction kit); sequencing template preparation kit and sequencing kit.

[0094] 2. Method

[0095] The four samples were built and processed on the computer according to the above experimental steps to obtain sample data, and then data analysis was performed.

Abstract

The invention discloses a quantitative method for free fetal DNA (deoxyribonucleic acid) proportion in maternal peripheral blood. DNA containing designed SNP (single nucleotide polymorphism) in plasma is extracted by the aid of a designed primer and loaded on a machine after a series of treatment, sequencing products are compared to a human genome sequence, and a basic group of each SNP site is determined. The concentration of the free fetal DNA is quantified by analyzing the proportion of the basic groups in the SNP. Induced errors can be corrected by the aid of a plurality of SNP sites, the SNP sites are simultaneously calculated by a statistics method, and calculated free fetal DNA concentration accuracy is improved.

Description

technical field [0001] The invention relates to the field of DNA quantification, in particular to a quantitative method for the proportion of fetal free DNA in the peripheral blood of pregnant women. Background technique [0002] Non-invasive prenatal genetic testing is to collect the peripheral blood (5ml) of pregnant women, extract the free DNA, use high-throughput sequencing technology, and combine biological information analysis to find that the fetus is suffering from chromosomal aneuploidy (21-trisomy, also known as Tang syndrome, 18-trisomy, 13-trisomy). Studies have found that from the 4th week of pregnancy, fetal cell-free DNA can be detected in the peripheral blood of pregnant women. As the gestational age increases, the content of fetal cell-free DNA also increases. After 12 weeks of pregnancy, by drawing the peripheral blood of the pregnant woman and extracting the fetal cell-free DNA, using the next-generation gene sequencing technology combined with bioinform...
