Embodiment 1

[0026] Embodiment 1: A kind of method of rapidly inducing and propagating the callus of the radicle grass radicle, the grass seed of the halophyte is placed in the ultra-clean workbench, first cleans the seed with sterile water, then processes 0.5- 1min, followed by cleaning the alcohol on the surface of the seeds with sterile water, and then treating with 20% sodium hypochlorite for 1.5-2min, and finally cleaning the sodium hypochlorite on the surface of the seeds with sterile water to obtain sterile seeds of Herba saline. The sterilized seeds are placed in the medium for cultivating sterile seedlings and cultivated for 10-15 days, and the roots of the sterile seedlings are obtained as explants for inducing callus. The medium is mainly composed of 1 / 2 Hoagland nutrient solution + 8g / L Composition of agar powder, PH=5.8; Aseptic seedling young roots are cut into small sections of about 0.5cm as callus-inducing materials, inoculated on callus-inducing medium and cultivated for 1...

Embodiment 2

[0028] Embodiment 2: A kind of method of rapidly inducing and propagating the callus of the radicle grass radicle, the grass seed of the halophyte is placed in the ultra-clean workbench, the seeds are first cleaned with sterile water, and then treated with 75% alcohol for 0.5- 1min, followed by cleaning the alcohol on the surface of the seeds with sterile water, and then treating with 20% sodium hypochlorite for 1.5-2min, and finally cleaning the sodium hypochlorite on the surface of the seeds with sterile water to obtain sterile seeds of Herba saline. The sterilized seeds are placed in the medium for cultivating sterile seedlings and cultivated for 10-15 days to obtain the roots of the sterile seedlings as explants for inducing callus. The medium is mainly composed of Hoagland nutrient solution and 6g / L agar powder Composition, PH=5.7; Transversely cut the young root of sterile seedlings into small sections of about 0.5 cm as callus-inducing materials, inoculate them on the ca...

Embodiment 3

[0031] A method for rapidly inducing and propagating the callus of the young roots of Herba haliflora, placing the seeds of Herba haliflora in a super-clean workbench, first cleaning the seeds with sterile water, and then treating them with 75% alcohol for 0.5-1min, followed by Clean the alcohol on the surface of the seeds with sterile water, then treat with 20% sodium hypochlorite for 1.5-2min, and finally clean the sodium hypochlorite on the surface of the seeds with sterile water to obtain sterile seeds of Herba saline, and sterilize the seeds Place in the medium for cultivating sterile seedlings and cultivate for 10-15 days to obtain the root system of sterile seedlings as explants for inducing callus. The medium is mainly composed of 1 / 2 Hoagland nutrient solution+6g / L agar powder, PH=5.8; Cut the young roots of sterile seedlings into small sections of about 0.5 cm as callus-inducing materials, inoculate them on the callus-inducing medium and cultivate them for 10 days to ...

Abstract

The invention relates to a method of inducing and culturing plant callus tissue and especially discloses a method of quickly inducing and breeding halogeton glomeratus radicle callus tissue. The method includes following steps: (1) obtaining sterilized explants; (2) performing callus tissue induction; and (3) performing callus tissue proliferation cultivation. Through subculture, a large amount of excellent callus tissue materials, which are white in color and crisp in structure, are obtained, and a problem of lignifications of root system is solved as well. The invention provides technical base for tissue culture of halogeton glomeratus radicle and cell suspension culture. The culture mediums used in induction and subculture of the callus tissue are all prepared with 2,4-D (2,4-dichlorophenoxyacetic acid) as an exogenous hormone, so that seedling culture, radicle callus induction and callus proliferation are all carried out under the same environment, thereby achieving a good effect. The halogeton glomeratus radicle callus tissue can be obtained simply, easily and high-effectively.

Description

technical field [0001] The invention relates to a method for inducing and cultivating plant callus, in particular to a method for rapidly inducing and proliferating callus of radicle grass roots. Background technique [0002] The culture of callus is an important stage in the process of plant tissue culture. Callus is a young parenchyma cell with potential differentiation ability, which can be further differentiated to form a complete plant. Different properties can also be obtained through callus culture. The callus can provide high-quality materials for the isolation, fusion or genetic transformation of protoplasts. Halogeton glomeratus is an annual herbaceous dilute halophyte of Chenopodiaceae, distributed in western Gansu, Qinghai, Xinjiang and Tibet; also found in Mongolia, Soviet Union, Siberia and Central Asia, growing at the foot of mountains, Gobi beach, suitable for growing and surviving in the desert. Halophytic grass absorbs water, high salinity, and nutrients ...
