Embodiment 1

[0033] Embodiment 1, promote the method for the accumulation of rosmarinic acid and salvianolic acid B in the hairy root of Salvia miltiorrhiza, carry out the following steps successively:

[0034] 1), initial cultivation of salvia miltiorrhiza hairy roots:

[0035] In an ultra-clean bench, inoculate 0.3 g of salvia miltiorrhiza hairy roots per 100 mL of 6,7-V medium; culture in the dark at 110 rpm and 25°C for 18 days;

[0036] 2), the preparation of folic acid solution:

[0037] Dissolve 0.2207g of folic acid in 10ml of sterile water, filter and sterilize with a 0.22μm sterile filter to obtain a folic acid solution, and store in a refrigerator at 4°C;

[0038] 3), the continued cultivation of salvia miltiorrhiza root:

[0039] After culturing the hairy root of Salvia miltiorrhiza for 18 days, folic acid solution was added to make the final concentration of folic acid in the 6,7-V medium 50 μM, and cultured (induced culture) in the dark at 110 rpm and 25° C. for 6 days.

Abstract

The invention discloses a method for promoting the accumulation of rosmarinic acid and salvianolic acid B in Salvia miltiorrhiza hairy roots. The method includes the following steps: 1) primary culture of the Salvia miltiorrhiza hairy roots: every 100 milliliters of 6,7-V culture medium is inoculated with 0.3 to 0.4 gram of the Salvia miltiorrhiza hairy roots, and darkness culture is carried out for 17 to 19 days under the culture conditions of 100 to 150 revolutions per minute and a temperature between 24 DEG C to 26 DEG C; 2) further culture of the Salvia miltiorrhiza hairy roots: a folic acid solution is then added so that the final folic acid concentration in the 6,7-V culture medium reaches between 45 microns and 55 microns, and darkness culture is carried out for 5 to 7 days under the culture conditions of 100 to 150 revolutions per minute and a temperature between 24 DEG C to 26 DEG C. The method provided by the invention has the advantage that folic acid is used for treating the Salvia miltiorrhiza hairy roots, so as to promote the accumulation of rosmarinic acid and salvianolic acid B, the secondary metabolites of the Salvia miltiorrhiza hairy roots.

Description

technical field [0001] The invention relates to a method for treating hairy roots of Salvia miltiorrhiza with folic acid, thereby promoting the accumulation of secondary metabolites rosmarinic acid and salvianolic acid B. Background technique [0002] Salvia miltiorrhiza is a plant of Labiatae; the roots and rhizomes of Salvia miltiorrhiza have the effects of promoting blood circulation and removing blood stasis, clearing away heat and calming nerves, nourishing blood and replenishing qi. Salvianolic acid is one of the active ingredients of Danshen, mainly including rosmarinic acid and salvianolic acid B. Due to its outstanding performance and non-toxic side effects in the treatment of cardiovascular diseases, the application and research of Danshen are becoming more and more extensive. [0003] Currently, the hairy root of Salvia miltiorrhiza can be cultured in 6,7-V medium. Generally, 0.15-0.2 g of fresh Salvia miltiorrhiza root is weighed and inoculated into 50 mL of 6,...
