AI Technical Summary

Benefits of technology

This patented technology helps reduce problems with producing certain compounds called teicophan by adding an ingredient like yeast extract or other substances into it beforehand when starting up new bacteria for use later on. By doing this, these techniques help prevent unwanted side reactions from affecting specific molecules involved in making Tecrolimus (a drug).

Problems solved by technology

Technological Problems This patented describes challengings associated with improving the efficiency at which tikilosan can produce certain substances like teiocyanolides called TICAOXINS. These new molecules were found earlier than previously identified because they had several advantages over existing ones. However, previous methods did not work well enough on this type of product without adding other ingredients. Therefore, improvements could lead to more effective ways to make teicopyranon from natural materials through gene manipulation techniques.

Embodiment approach 1

[0023] Embodiment 1: No fed-batch fermentation

[0024] (1) Seed culture: inoculate the swimming actinomycetes into the seed culture medium, cultivate in a 30L test tank, and the culture period is 50±16h;

[0025] Seed medium formula (parts by mass): soybean meal powder 2.0, yeast powder 0.5, peptone 0.5, glucose 2.5, starch 2, sodium chloride 0.4, potassium dihydrogen phosphate 0.02, calcium carbonate 0.3;

[0026] Cultivation conditions: temperature 28℃, ventilation volume v:v 1:1.5, tank pressure 0.05Mpa, rotating speed 300rpm, initial pH value 7.2;

[0027] (2) Fermentation culture: Inoculate the cultured seed liquid into the fermentation medium according to the inoculation amount of 6%, and cultivate it in a 50L test tank. The culture period is 120±60h;

[0028] Fermentation medium formula (parts by mass): starch 3, magnesium sulfate heptahydrate 0.45, calcium carbonate 0.6, sodium chloride 0.08, peptone 0.1, yeast powder 1, soybean meal 1.5, valine 0.1.

[0029] Culture conditions:...

Embodiment approach 2

[0030] Embodiment 2: No fed-batch fermentation, pH adjustment

[0031] Repeat embodiment 1 in Example 1, adjust the pH value with phosphoric acid or ammonia water, so that the pH value is maintained at 6.6-7.4.

Embodiment approach 3

[0032] Embodiment 3: Add carbon and nitrogen sources to adjust pH

[0033] Repeat embodiment 1 in Example 1 to detect the mass percentage concentration of total sugar, amino nitrogen and pH value. When the mass percentage concentration of total sugar is less than 1.2%, add carbon source; when the concentration of amino nitrogen is less than 20g / L When, add a nitrogen source; adjust the pH value with phosphoric acid or ammonia water to keep the pH value at 6.6-7.4.

[0034] In fed-feed fermentation, the carbon source is an aqueous solution of glucose, and the mixing ratio w / v of glucose and water is 40:100;

[0035] In the fed-batch fermentation, the nitrogen source is an aqueous solution of soybean powder, and the mixing ratio of soybean powder and water w / v is 30:100.

[0036] The teicoplanin fermentation conditions of the embodiments 1-3 are shown in Table 1:

Abstract

The invention provides a supplemented culture medium of teicoplanin, which is capable of improving the output of teicoplanin and a method for producing the teicoplanin. The supplemented culture medium of the teicoplanin comprises one or more of a carbon source, a nitrogen source and a pH value regulator, wherein the carbon source comprises one or more of glycerol, glucose, starch, oat flour and lactose; the nitrogen source comprises one or more of corn gluten meal, cottonseed fine lour, soybean meal, peptone and yeast powder; and the pH value regulator comprises one or two of phosphoric acid and ammonia water. The method for producing the teicoplanin specifically comprises a step of adding the supplemented culture medium into a fermentation culture medium during fermentation, wherein the supplemented culture medium comprises one or more of the carbon source, the nitrogen source and the pH value regulator. The invention has the beneficial effects that the supplemented culture medium is added into the fermentation culture medium during fermentation to reduce factors inhibiting secondary metabolic pathways for producing the teicoplanin, and the fermentation output of the teicoplanin can be improved.
