Problems solved by technology

Although the pathogenic bacteria of this model are the same as the most common pathogenic bacteria of human BBF-associated sinusitis, the results are relatively reliable, but there are still the following defects: 1. The process is complicated, requiring general anesthesia and endotracheal intubation for animals Assisted breathing; 2. The operation is difficult, and the sheep's middle turbinate and anterior ethmoid sinus need to be removed under nasal endoscopy; then, fluorescein is injected through the frontal sinus anterior wall drilling, and the position of the frontal sinus opening is confirmed under nasal endoscopy And be completely blocked; 3, using sheep as a model animal is expensive and inconvenient to operate, not suitable for scientific research that requires a large sample size

[0046] (1) Screening for BBF-positive Staphylococcus aureus:

[0047] Collect clinical specimens, sort and identify Staphylococcus aureus by conventional methods, pick a single colony and inoculate into TSB culture solution (TSBg) containing 0.5% glucose, shake overnight at 37°C and dilute 200 times with TSBg, add polystyrene material In a 96-well plate, 200uL per well, cultured at 37°C for 24 hours, washed with PBS to remove planktonic bacteria, fixed with methanol for 15 minutes, air-dried for 30 minutes after aspirating the methanol, added 200uL of 1% crystal violet solution, stained for 15 minutes, and rinsed thoroughly with PBS Add 200 uL of 95% ethanol to each well of the unbound crystal violet staining solution after drying, place it on a shaker at room temperature for 1 hour, and measure the absorbance (OD value) at 490 nm with a microplate reader. OD value ≥ 0.12 is considered to be a BBF-positive strain, and the larger the OD value, the stronger the ability to form B...

Abstract

The invention belongs to the biomedical field and relates to a model making method of a bacterial biofilm (BBF)-related nasosinusitis animal model. By the use of clinical strains of BBF positive staphylococcus aureus and an animal model and through indwelling absorbable gelatin sponge, a BBF-related nasosinusitis disease model is established. Through scanning electron microscope detection, the result shows that three-dimensional structured mature BBF formed after bacterial adhesion exist on the surface of mucous membrane of the animal model, and it is verified that modeling of the BBF-related nasosinusitis disease model is successful. The method has the following characteristics: pathogenic bacteria is the same with the most common pathogenic bacteria in human BBF-related nasosinusitis; the model making method is simple, has high operationality and requires no special surgical instruments; influence on normal structures and functions is little; the indwelling absorbable gelatin sponge is used as a carrier for bacteria proliferation, success rate of modeling is high, and results are stable; and the obtained BBF-related nasosinusitis disease model is more similar to human related diseases.

Description

technical field [0001] The invention belongs to the field of biomedicine, and relates to a modeling method for establishing an animal disease model of bacterial biofilm-associated sinusitis. Background technique [0002] Research reports that bacterial biofilm (bacterial biofilm, BBF) is a protective growth mode of bacteria. Due to the special structure of BBF, antibiotics cannot effectively kill the bacteria inside, and at the same time, the antibodies produced by the body not only Bacteria cannot be killed, but immune complexes are formed on the surface of BBF, resulting in the persistence of inflammatory reactions. Chronic sinusitis is a common disease in the Department of Otorhinolaryngology, about 10% of the patients have not been able to obtain the ideal clinical treatment effect, and this part of the patients is usually called refractory sinusitis. Studies have found that the positive rate of bacterial biofilm on the nasal mucosa of patients with refractory sinusitis...
