Embodiment 1

[0040] Reference figure 1 An electrochemical sensor provided by the present invention includes a glassy carbon electrode as a working electrode. A graphene layer and a nano-gold layer are deposited on the surface of the reaction end of the glassy carbon electrode, and the nano-gold layer is self-assembled with sulfhydryl modified available T-Hg 2+ -T mismatch to form a mercury-specific oligonucleotide probe of double-stranded DNA structure (ie figure 1 S1). In the above electrochemical sensor, the nucleotide sequence of the mercury-specific oligonucleotide probe S1 after sulfhydryl modification is the nucleotide sequence of SEQ ID No. 1, specifically: 5'-SH-(CH 2 ) 6 -GTG TTT CTC A-3’.

Abstract

An electrochemical sensor for detection of trace mercury in a water body comprises a glassy carbon electrode which can be used as a working electrode in a three-electrode system; the surface of a reaction end of the glassy carbon electrode is deposited with a graphene layer, the surface of the graphene layer is deposited with a nano gold layer, the surface of the nano gold layer is self-assembled with a mercury specific oligonucleotide probe which is modified by a mercapto group and can form a double-stranded DNA structure by using T-Hg<2+>-T mismatching. With adopting of the mercury specific oligonucleotide probe for self-assembly on the surface of the reaction end, a stereoscopic response domain which can specifically capture mercury ions is built; and the electrochemical sensor is used for high-efficiency detection of trace mercury in the water body, and has the advantages of simple structure, practicality, convenient manufacture and use, strong environmental impact resistance, good selective specificity, ultra sensitivity and the like.

Description

Technical field [0001] The invention relates to the technical field of detection of mercury ion content, in particular to an electrochemical sensor for detecting mercury ions, and a preparation method and application thereof. Background technique [0002] Mercury and its compounds are very harmful to the environment and human health. It is of great practical significance to efficiently and sensitively monitor and analyze the mercury content in food, water bodies, and the environment closely related to human life. [0003] Traditional mercury analysis methods include coupled plasma mass spectrometry, atomic emission spectroscopy, atomic fluorescence spectroscopy, ion chromatography, etc. These analysis methods all have cumbersome pretreatment steps, expensive equipment, and complex operations, which limit the traditional methods in environmental sample traces. Application in the measurement of mercury. In recent years, as a new type of detection method, sensor technology is develop...
