Problems solved by technology

[0003] In recent years, researchers have conducted a lot of research on the tissue culture rapid propagation technology of Dendrobium candidum, but they mainly focus on the proliferation and differentiation of protocorms, and there are not many studies on the rapid propagation of Dendrobium candidum in vitro using stem segments as materials.

Embodiment 1

[0013] The rapid propagation method of the Dendrobium officinale of the present embodiment may further comprise the steps:

[0014] (1) Adventitious bud induction: Take the young stems of Dendrobium candidum in the same year, cut them into small stems with a length of 1-1.5cm and 1-2 axillary buds, soak them in 75% alcohol for 30s after washing, and then Disinfect with 0.1% mercuric chloride for 10 minutes, and then rinse with sterile water for 5 times;

[0015] Cultivate the sterilized dendrobium stem section in the induction medium for 30 days. The induction medium is: MS basic medium, 0.1mg / L naphthaleneacetic acid, 0.5mg / L 6-benzylaminoadenine, 3% sucrose, 0.8 % agar;

[0016] (2) Proliferation of adventitious buds: After the induction, cut off the new shoots that grow from the stem segments at the internodes, inoculate them in the induction medium for subculture and proliferate for 30 days, and obtain clustered buds at the cuts;

Abstract

The invention relates to a rapid propagation method of dendrobium officinale. The method comprises the steps as follows: (1) adventitious bud induction: culturing disinfected dendrobium nobile stems in an induction medium for 28-30 d, wherein the induction medium comprises an MS (murashige and skoog) minimal medium, 0.1-0.15 mg / L of 1-naphthylacetic acid, 0.5-1.0 mg / L of 6-benzylaminopurine, 3% of sucrose and 0.8% of agar; (2) multiplication of adventitious buds: after induction, cutting off buds growing at internodes of the stems, and inoculating the buds in the induction medium for subculture multiplication for 30-32 d to obtain cluster buds at cuts; (3) strong seedling and rooting transplant: cutting the cluster buds into single seedlings, transferring the single seedlings in a strong seedling medium for strong seedling culture for 23-25 d, and performing rooting culture for 30-32 d when each bud is 2-3 cm high and has 2-3 blades, wherein the strong seedling medium comprises an MS minimal medium, 3% of sucrose and 0.7% of agar, and the rooting medium comprises a 1 / 2 MS minimal medium, 0.3-0.5 mg / L of 1-naphthylacetic acid and 0.3-0.5% of activated carbon. According to the dendrobium officinale propagated with the method, heights of the cluster buds are above 3.9 cm, the number of roots is above 4, and lengths of the roots are above 1.5 cm.

Description

technical field [0001] The invention relates to a cultivation technology of dendrobium candidum, in particular to a rapid propagation method of dendrobium candidum. Background technique [0002] Dendrobium candidum, also known as black knot grass, commonly known as Tiepi Fengdou, is a perennial evergreen herb of the genus Dendrobium orchidaceae, and is a precious medicinal material. In recent years, studies have found that Dendrobium contains polysaccharides, alkaloids, phenanthrene and erianin and other compounds, as well as 7 kinds of essential amino acids and various trace elements. Modern pharmacology shows that Dendrobium has anti-tumor, anti-aging, and immune-enhancing functions, and has obvious therapeutic effects on cardiovascular, digestive system, respiratory system and ophthalmic diseases. Due to the medicinal value of Dendrobium candidum with many unique curative effects, it is called "Jiancao of life-saving" among the people, and "the giant panda of the medicin...
