Abstract

The invention discloses a method for obtaining piperine via direct column chromatography, which belongs to the technical field of medicine preparation. The method comprises the following steps of: taking 1 part by weight of pepper powder passing through a 100-mesh sieve, and 0.8-1.2 parts by weight of silica gel or aluminium oxide with a particle size of 80-100 meshes, uniformly mixing the two raw materials and grinding; spreading the obtained product in a glass chromatography column filled with 0.8-5.0 parts by weight of silica gel or aluminium oxide with a particle size of 80-100 meshes, eluting by taking petroleum ether-acetone or petroleum ether-ethyl acetate or chloroform-acetone or chloroform-ethyl acetate in a volume ratio of 5: (0.5 to 2.0) as a mobile phase, and collecting fractions; and performing reduced pressure concentration on the fractions containing piperine, and then crystallizing by methanol-water in a volume ratio of (7.8 to 8.2): 2 to obtain faint yellow piperine massive crystals. Compared with the prior art, the method disclosed by the invention has the advantages that extraction and purification processes of piperine are optimized, the operation steps of solvent extraction and the types of the used solvents are reduced, and the consumption of the solvents is decreased; and moreover, the obtained product is stable in quality, high in productivity, low in cost, and low in time consumption.
