Embodiment Construction

[0062] The agents and various hormones used in the minimal medium in this example were all purchased from Sigma Company.

[0063] A method for in vitro culture and rapid propagation of Rock Chinese cabbage, wherein the explants of Rock Chinese cabbage are subjected to primary culture, proliferation culture, strong seedling culture and rooting culture to obtain regenerated plants, and then transplanted, comprising the following steps:

[0064] First: the genus Petracea mainly contains three types of substances: phenols, flavonoids and polysaccharides, etc., which can easily cause browning of plant materials in in vitro culture, so the explants should not be trimmed too small during disinfection. Rock cabbage has thick rhizomes and is buried deep in the soil for a long time. There are many chances of infecting various exogenous microorganisms, so it is very difficult to obtain completely sterile materials through disinfection. Through repeated experiments, the present invention ...

Abstract

The invention relates to a method for performing in-vitro culturing and rapid propagating on bergenia crassifolia. The method comprises the following steps of: selecting a fresh rhizome with terminal buds as a sterile exophyte, and inoculating the sterile exophyte to a primary culture medium to carry out primary culture; transferring a primarily-cultured aseptic seedling to a proliferation culture medium, wherein the proliferation culture medium comprises 0.5-3.5 mg.L<-1> of MS (Murashige and Skoog)+BA (Butyl Acylate), 0.2-1.5 mg.L<-1> of NAA (Naphtyl Acetic Acid), 0.5-5.0 mg.L<-1> of VC (Vitamin C), 25-35g.L<-1> of sucrose and 4-84.L <-1> of agar; inducing adventitious buds; slitting the induced cluster buds and transferring the slit cluster buds to a strong seedling culture medium for culturing the strong seedling; slitting test-tube seedling and transferring the slit test-tube seedling to a rooting medium, wherein the rooting medium comprises 1.5-4.5 mg.L<-1> of 1 / 2MS+NAA, 0.5-2.5 mg.L<-1> of IBA (Indole Butyric Acid), 0.5-5.0 mg.L<-1> of VC, 15-25.gL<-1> of sucrose and 4-8g.L<-1> of agar; cleaning and transferring the rooted test-tube seedling to a substrate for planting. The method can be used for increasing a yearly propagation coefficient to 1:600000 and reducing the cost to 2-3 yuan per plant, and is free of season limitation and easy in environment control, so that variant plants are reduced, the quality of the seedling is high, and the consistency is good.

Description

technical field [0001] The invention belongs to the technical field of rapid plant propagation methods, in particular to in vitro culture and rapid propagation of rock cabbage. Background technique [0002] rock cabbage ( Bergenia crassifolia (Linn.) Fritsch) is a rock cabbage of the family Saxifragaceae, which is a perennial evergreen herb. It is a larger species of plants in the same genus. The domestic wild species are only found in the rock crevices under the bushes at an altitude of about 2000 m in the Altai Mountains of Xinjiang. There are also a small amount of distribution in Mongolia, Russia, and North Korea. It blooms very early and enters winter. The leaves are red-green or purple-red. The flowers and leaves are beautiful and have high ornamental value. The whole plant contains petracenin, which is a special drug for treating respiratory diseases. Petracein exists in five families and nine genera of plants in my country, and it is also found in plants in Italy, ...
