Embodiment Construction

[0016] In order to make the object, technical solution and advantages of the present invention clearer, the implementation manner of the present invention will be further described in detail below in conjunction with the accompanying drawings.

[0017] A blocking solution for biochips used for detecting multiple proteins, the blocking solution is polyvinylidene fluoride blocking solution. That is, the polyvinylidene fluoride blocking solution is used as a blocking solution in biochips for detecting various proteins.

[0018] The polyvinylidene fluoride blocking solution of the present invention was purchased from NOF Corporation, and its product catalog name is "blocking reagent N101", wherein the main component is PVDF. By adopting the blocking solution of the present invention, non-specific free binding and cross-reaction are eliminated, and false positives are avoided.

Abstract

The invention discloses a confining liquid of a biochip for detecting various proteins. The confining liquid is a polyvinylidene fluoride confining liquid. The confining liquid is 50-100 percent solution obtained by diluting phosphate buffer. The invention provides a confining method of the biochip for detecting various proteins. The confining method comprises the following steps of: firstly, adding a primary antibody and adding the confining liquid after reaction; secondly, carrying out cryopreservation on the mixed solution at the temperature of 40DEG C below zero for two hours; and thirdly, carrying out freeze drying on the mixture at the temperature 80DEG C below zero in a vacuum state. According to the confining liquid and the confining method disclosed by the invention, the polyvinylidene fluoride confining liquid is used as the confining liquid in the biochip for detecting various proteins; and when the biochip is used for detecting various proteins, the non-specific free combination and cross reaction can be eliminated and false positive is avoided.

Description

technical field [0001] The invention relates to the field of biological detection, in particular to a blocking liquid and a blocking method of a biochip used for detecting various proteins. Background technique [0002] Since Engvall and Perlmann published an article on enzyme-linked immunosorbent assay (ELISA, Enzyme Linked Immunosorbent Assay) for the quantitative determination of immunoglobulin G (IgG) in 1971, this enzyme-labeled antibody technology began to be used for antigen localization Developed into a method for the determination of trace substances in liquid specimens. The basic principle of the antigen-localized enzyme-labeled antibody technology is: ① Make the antigen or antibody bind to the surface of a certain solid phase carrier and maintain its immunological activity. ②The antigen or antibody is connected with an enzyme to form an enzyme-labeled antigen or antibody, which not only retains its immune activity, but also retains the activity of the enzyme.
