Embodiment 1

[0069] Preparation of composition doses of tramadol hydrochloride and celecoxib

[0070] Preparation of racemic tramadol hydrochloride and celecoxib at different molar ratios (1:1, 1:3 and 3:1) of (racemic)-tramadol·HCl:celecoxib combination. All drugs and combinations were dissolved in distilled water containing 0.5% hydroxypropylmethylcellulose and administered at 10 ml / kg per rat via intraperitoneal (i.p.) route. Table 1 lists the different ratios prepared at various concentrations.

[0073] Effects of thermal hyperalgesia in a postoperative pain model in rats

[0074] The aim of this study was to evaluate the elimination of the combination comprising tramadol / celecoxib, in particular at different molar ratios (1:1, 1:3 and 3:1), in a model of postoperative pain following paw incision in rats. Analgesic efficacy and potency of a combination of tramadol hydrochlor...

Abstract

The present invention relates to pharmaceutical compositions comprising tramadol and celecoxib and their uses as medicaments or analgesics, more particularly for the treatment of severe to moderate pain with an inflammation component.

Description

technical field [0001] The present invention relates to a pharmaceutical composition comprising tramadol and celecoxib, and its use as a medicament or an analgesic, more particularly for the treatment of severe to moderate pain. Background technique [0002] Pain is a complex response that is functionally classified into sensory, autonomic, motor, and affective components. The sensory aspect includes information about the location and intensity of stimuli, while the adaptive component can be viewed as the activation of endogenous pain modulation and motor planning in response to escape. The affective component appears to include the assessment of pain as unpleasant and stimuli-threat as well as negative emotions triggered by memory and the painful stimulus environment. [0003] In general, pain conditions can be divided into chronic and acute. Chronic pain includes neuropathic pain as well as chronic inflammatory pain, such as arthritis, or unexplained pain, such as fibrom...
