Embodiment 1

[0044] Pass 2g of selenium-enriched yeast with a selenium content of 2000ppm through a 200-mesh sieve to remove impurities in the yeast. The selenium-enriched yeast is mixed with a sodium hydroxide solution having a pH of 13.0 in a mass ratio of 1:5. The mixed solution was placed in a shaker at 250r / min and 35°C for 8 hours, and the mixed solution was taken out at two time points of 3 hours and 6 hours, and was ultrasonicated for 30 minutes each time and then returned to the shaker. After the shaking is completed, the mixed solution is centrifuged in a centrifuge with a rotating speed of 10000r / min for 30 minutes to separate impurities and extract the supernatant. The pH of the supernatant was adjusted to 7 with dilute hydrochloric acid to obtain a selenoprotein-containing solution. According to the selenium content in the extraction supernatant, calculate the total selenium extraction rate.

Embodiment 2

[0046] Pass 2g of selenium-enriched yeast with a selenium content of 2000ppm through a 200-mesh sieve to remove impurities in the yeast. The selenium-enriched yeast is mixed with a sodium hydroxide solution having a pH of 13.3 in a mass ratio of 1:5.

Embodiment 3

[0048] Pass 2g of selenium-enriched yeast with a selenium content of 2000ppm through a 200-mesh sieve to remove impurities in the yeast. The selenium-enriched yeast is mixed with a sodium hydroxide solution having a pH of 14.0 in a mass ratio of 1:5. The mixed solution was placed in a shaker at 250r / min and 35°C for 8 hours, and the mixed solution was taken out at two time points of 3 hours and 6 hours, and was ultrasonicated for 30 minutes each time and then returned to the shaker. After the shaking is completed, the mixed solution is centrifuged in a centrifuge with a rotating speed of 4000r / min for 30 minutes to separate impurities and extract the supernatant. The pH of the supernatant was adjusted to 7 with dilute hydrochloric acid to obtain a selenoprotein-containing solution. According to the selenium content in the extraction supernatant, calculate the total selenium extraction rate.

Abstract

The invention discloses a method for extracting selenium-containing protein from selenium-enriched yeast, which is characterized by comprising the following steps: (1) choosing the selenium-enriched yeast as raw materials, using a sodium hydroxide or potassium hydroxide solution with potential of hydrogen (pH) of 11 to 14 to dissolve the selenium-enriched yeast to obtain mixed liquid A; (2) placing the mixed liquid in a vibration machine of 250 r / minute at the temperature of 35 DEG C to be vibrated for 8 hours to 12 hours, and performing ultrasound on the mixed liquid for 30 minutes for twice in vibration processes; and (3) arranging the mixed liquid in a centrifugal machine of 4,000 to 20,000 r / min for 30 minutes after vibration is finished to extract supernatant, and adjusting pH of the supernatant to 7 to obtain a selenium-containing protein solution. The method improves wall broken rate and protein dissolving rate of yeast cells, extraction rate of selenium in the selenium-enriched protein solution is more than 70% when the selenium-enriched solution is not filtered and purified, and selenium-containing protein with organic selenium content more than 95 % can be obtained after the selenium-enriched solution is filtered and purified.

Description

technical field [0001] The invention provides a method for extracting selenoproteins with selenium-enriched yeast, which belongs to the preparation and processing of food, and belongs to A23L according to IPC classification. Background technique [0002] Selenium is an essential trace element for animals and the human body. Selenium deficiency will seriously affect human health. Selenium can improve human immunity, scavenge free radicals, protect optic nerve, heart and liver, etc., and can prevent diabetes, cardiovascular and cerebrovascular diseases, liver diseases, cataracts, arthritis, Keshan disease, Kaschin-Beck disease and other major diseases. The form of selenium is often divided into organic selenium and inorganic selenium. Inorganic selenium includes selenite, selenate, selenide, and elemental selenium, and organic selenium includes selenomethionine, selenocystine, and selenocysteine. Inorganic selenium is difficult to be directly absorbed and utilized by animals...
