Problems solved by technology

These utensils are relatively hard, which is easy to injure the parent fish and affect the subsequent spawning, especially when separating the egg mass, it is easy to pierce the egg membrane, resulting in the failure of hatching

Embodiment Construction

[0010] The invention utilizes a commercially available disposable inoculation loop to collect and separate fertilized eggs, and the inoculation loop is made of soft and elastic engineering plastic material. The fertilized eggs of medaka fish are about 1mm in size, so it is advisable to choose an inoculation loop with a hole diameter slightly larger than the egg diameter when purchasing.

[0011] The invention is equipped with auxiliary tools such as a small net, a glass petri dish, and a large-mouth straw. The soft small net is used to fish for broodstock; fertilized eggs are separated in a glass petri dish with a size of 9 to 12 cm;

[0012] The operation method is carried out according to the following steps (see accompanying drawings 1 and 2):

[0013] Within 1 hour to 6 hours after spawning, fish the female fish with eggs hanging on the abdomen to the water surface with a soft small net (do not leave the water surface);

[0014] Use a disposable inoculation loop 2 to remo...

Abstract

The invention discloses a method for collecting and separating medaka zygotes, and relates to the technology of fish zygote separation. In order to solve the technical problems, the invention provides the method for collecting and separating the medaka zygotes for preventing egg membranes from being punctured. The invention adopts the technical scheme that the method adopts a soft small brail, disposable engineering plastic inoculation cups, a large-mouth suction tube and culture dishes. The method is characterized by comprising the following steps of: fishing female fishes of which bellies are hung with eggs within 1 to 6 hours after laying eggs to the water surface by using the soft small brail, taking down the egg blocks by using the disposable inoculation cup, and transferring the taken-down egg blocks to the glass culture dish filled with Ringer's culture solution to perform separation; and mutually stretching the inoculation cups held in the left and right hands to break the egg silks and separate the zygotes, sucking the separated single eggs to the other culture dish filled with the Ringer's culture solution by using the large-mouth suction tube to perform hatch, and controlling the hatching temperature to be between 25 and 26 DEG C. The method is used for embryo observation and experiments.

Description

technical field [0001] The invention belongs to fish fertilized egg separation technology. Background technique [0002] Medaka (Oryzias latipes) is a small fish native to East Asia that is easy to keep. The adult fish is about 3cm long and is one of the smallest known vertebrates. Compared with the experimental animals zebrafish and mice, the generation is shorter, 2 to 3 months. In an artificial environment, medaka can spawn every day of the year, and the spawning time can be controlled by controlling the light time. The transparent fertilized egg makes it very convenient for embryo observation and micromanipulation. Because spawning is controllable, medaka, as a representative of vertebrates, was sent into space in 1994, completing the entire development process from fertilization to individuality, realizing the real "space breeding". Japan started research on medaka at the beginning of the last century, and it has been widely used in physiology, ecology, endocrinolog...
