The method is that ISSR-SCARs markers and an identification method for identifying dioscorea collettii hook.f. are found on the basis of the processes of the dioscorea collettii hook.f. and dioscorea hypoglauca palib which is the orthodox origin of Chinese traditional medicine rhizoma dioscoreae hypoglaucae such as total DNA extraction, PCR amplification, primer screening ISSE-SCARs amplification, testing and analysis, etc. Using the primers tdisclosed by the invention to carry out the PCR reaction can obtain amplified fragments with 960bp length, which do not have in the dioscorea hypoglauca palib, from the dioscorea collettii hook.f.
