[0019] 1. Preparation of DCIK cell culture supernatant

[0020] The DCIK cells matured on the 9th day after DC and CIK cells were mixed and induced, and then entered the stage of mass expansion and preparation. When the cell concentration reaches 1~5×10 6 After cells / ml, the uniformly mixed DCIK cells and the culture supernatant were divided into two, and an equal volume of medium was added to continue the culture. After the DCIK cells matured in culture, centrifuge at 2000rpm for 10min to take the supernatant.

[0021] In order to determine the number of culture days of the supernatant with the best tumor killing activity, a small amount of DCIK cell culture supernatants on days 9, 12, 16, 20, 23, and 26 were collected for a small amount of pre-experiment and applied to in vitro culture melanoma B16 cells to detect its growth inhibitory effect. The experimental results found that the tumor cell killing rates of supernatants of these different days were 10%, 50%, 50%, 60%, ...

Abstract

The invention relates to supernatant cultivated by DCIK cells and an application thereof. The supernatant and a preparation of freeze-dried powder prepared by frozen, dried and concentrated supernatant have good anti-tumor activity in tumor therapy. The supernatant cultivated by DCIK cells contains a series of mixtures of cytokines with biological activity which directly act on cell lines cultivated in vitro, thereby causing the forms of the cell lines such as mice melanoma B16 and THP-1 etc. to be affected and producing growth inhibition. The anti-tumor in vivo experiment shows that the supernatant cultivated by DCIK cells and the preparation of freeze-dried powder thereof can obviously inhibit the growth of the mouse melanoma B16 inoculated subcutaneously without obvious side effect. The invention fully proves the prospects of application and research of supernatant cultivated by DCIK cells, and paves the new way for the research of anti-tumor drugs.

Description

technical field [0001] The invention relates to a DCIK cell culture supernatant and application thereof. Background technique [0002] Current clinical treatments for malignant tumors mainly include surgery, chemotherapy and radiotherapy. In recent years, with the rapid development of biomedicine, cellular immunotherapy as an adjuvant therapy has begun preliminary clinical application and achieved certain curative effects. Among them, DCIK cells are newly reported immune effector cells with good anti-tumor effect. [0003] DCIK cells are the effector cell population obtained by the co-culture of dendritic cells (DC) and cytokine-induced killer cells (CIK). Studies have shown that the proliferation, cytotoxic activity, and antigen specificity of DCIK effector cells are significantly higher than those of CIK cells. enhanced. At present, DCIK cells have achieved good curative effect in the results of clinical phase II trials, and the treatment effect on common tumors is rema...
