Embodiment Construction

[0042] Such as figure 1 Shown is a schematic diagram of a biosensitive film field effect sensor with a stacked structure. A P-type silicon material 3 is used to manufacture a MOS type field effect transistor by a semiconductor manufacturing process. The field effect transistor includes a source 1 , a drain 2 , and a gate made of non-metallic insulating material silicon nitride 4 . Silanize the sensitive gate silicon nitride 4, immobilize protein A5 on the surface of silicon nitride 4 by adsorption method, dry at room temperature, seal with bovine serum albumin (BSA), wash with phosphate buffer solution (PBS) and wait for drying. This forms a silicon nitride-protein A composite sensitive film. Then, the antibody 6 of TRAIL receptor was immobilized on the protein A5 by adsorption method, dried at room temperature, washed with phosphate buffer solution (PBS) and dried for later use. So far, a silicon nitride-protein A-TRAIL receptor antibody has been prepared as a biologically...

Abstract

The present invention discloses the preparation process of biosensitive TRAIL antibody membrane. The preparation process includes the following steps: A. semiconductor manufacture process of making MOSFET including source, drain, and grid of non-metal insulating material SiN with P-type silicon material; B. silanizing sensitive SiN grid; C. fixing protein A onto the surface of SiN via adsorption, drying at room temperature, closing with ox serum protein, cleaning with buffering phosphate solution and drying to form composite SiN-protein A sensitive membrane; and D. fixing antibody to TRAIL receptor via adsorption onto protein A, drying at room temperature, cleaning with buffering phosphate solution and drying to form the superposed biosensitive SiN-protein A- TRAIL receptor antibody membrane. The present invention also relates to multipath TRAIL receptor detection system with the membrane and method of detecting four TRAIL receptors of tumor cell apoptosis factor.

Description

Background technique [0001] Apoptosis is one of the most important physiological and pathological phenomena in living organisms. TRAIL (TNF-relatedapoptosisindecingligand) is a new member of the tumor necrosis factor TNF (Tumomecrosisfactor) family, and its greatest biological feature is selective cytotoxicity, that is, it only induces the apoptosis of tumor cells, transformed cells or virus-infected cells, and does not cause normal apoptosis. This feature has potential and broad application prospects in tumor therapy. [0002] At present, there are two methods for detecting the four receptors of TRAIL. One is to use the method of flow cytometry to detect in the laboratory. Each patient's sample needs to be individually labeled with four antibodies and operated separately. The operation process is complicated, and it is easy to cause waste of reagents and consumables. The method of flow cytometry is only suitable for the research of a small number of specimens in the labor...
