Abstract

The invention discloses a method for directly detecting H7 subtype avian influenza virus without amplification based on CRISPR-Cas13a. The method comprises the following steps: (1) extracting RNA (Ribonucleic Acid) of a sample; (2) preparing a CRISPR-Cas13a detection and analysis system; (3) adding RNA (Ribonucleic Acid) of a sample to be detected into a detection and analysis system; and (4) reacting for 30-60 minutes under the isothermal condition of 37 DEG C, and detecting a fluorescence signal released by the CRISPR-Cas13a in an attached cleavage manner in real time. The invention discloses a method for directly and rapidly detecting RNA (Ribonucleic Acid) of an H7 subtype avian influenza virus by utilizing a CRISPR-Cas13a (Clustered Regularly Interspaced Short Palindromic Repeats-Cas13a) detection system without pre-amplification of a viral genome. Compared with the conventional CRISPR detection method, the method disclosed by the invention is good in sensitivity, strong in specificity and simpler and more convenient to operate, the reaction time is greatly shortened, and rapid, accurate and low-cost screening can be carried out on the H7 subtype avian influenza virus.
