[0068] Through directed mutagenesis and cloning of nucleic acid fragments, a recombinant pestivirus based on the cDNA clone of the KSPV isolate "Alfort-Tübingen" (genotype 2.3) was generated (Meyers et al., 1996, J Virol. 70(3): 1588-95 ). Here, the complete E of KSPV Alfort-Tübingen rns The coding sequence was substituted by the corresponding regions of pronghorn pestivirus, rat pestivirus, and combinations consisting of rat pestivirus and pronghorn pestivirus. Starting from the RNA of a pronghorn pestivirus isolate or from a nucleic acid synthesized in rat pestivirus based on the published sequence, for this purpose the corresponding E rns Gene fragment. About 20 nucleotides complementary to sequence fragments immediately upstream and immediately downstream of the genomic region to be replaced were fused at both ends of the amplicon via primers. Via the "target sequence-primed plasmid amplification" method, the pronghorn or rat pestivirus sequences were introduced into a ...

Abstract

In a first aspect, the present invention relates to a genetically modified pestivirus, which is characterized by the fact that it is modified in such a way that at least the gene segment coded for theEms peptide originates from a distantly related pestivirus or from a plurality of pestiviruses that are distantly related to the genetically modified pestivirus. In a further aspect, the invention relates to an infected host cell or its cell culture supernatant containing a pestivirus genetically modified according to the invention. The invention is also relates to a vaccine and to its use and tothe use of the genetically modified pestivirus as a vaccine, in particular as a vaccine for animals and as a vaccine against classical swine fever (CSF), bovine virus diarrhoea (BVD), border disease(BD) and other illnesses caused by pestiviruses. The invention further relates to a recombinant chimaeric Ems peptide, and to detection systems for detecting pestiviruses or antibodies induced thereby, in particular for differentiating between vaccinated animals and animals after a natural infection with pestiviruses. Finally, the invention relates to a method for determining the extent to which an animal has been vaccinated with a vaccine according to the invention, and also to diagnostic kits for this purpose and to methods for controlling an infection with a pestivirus in a population of animals from the order of Artiodactyla.

Description

technical field [0001] In a first aspect, the invention relates to a genetically modified pestivirus, characterized in that said pestivirus is modified such that at least for E rns The peptide-encoding gene segment is derived from one or more pestiviruses distantly related to the genetically modified pestiviruses. In another aspect, an infected host cell or a cell culture supernatant thereof comprising a pestivirus genetically modified according to the invention is described. Furthermore, the invention relates to a vaccine and its use as well as the use of a genetically modified pestivirus as a vaccine, in particular as a vaccine for animals and as a vaccine against classical swine fever (KSP), bovine viral diarrhea (BVD), border disease (BD) and other vaccines for diseases caused by pestiviruses. Furthermore, a recombinant chimeric E rns Peptides and a detection system for the detection of pestiviruses or antibodies induced therefrom, in particular for differentiating vacc...
