Abstract

The invention provides an electrochemical luminescence immunoassay method which comprises the following specific steps: fully reacting a sample, a biotinylated cortisol specific antibody and a ruthenium pyridine labeled cortisol derivative, and then adding streptavidin-coated magnetic bead particles for incubation to obtain an antigen-antibody magnetic particle complex; sucking the antigen-antibody magnetic particle complex into a measuring cell, adding tripropylamine, applying voltage to generate ECL reaction, enabling the antigen-antibody magnetic particle complex to emit chemiluminiscence,and measuring luminous intensity through a photomultiplier so as to obtain the concentration of cortisol. The method is convenient and rapid, and accurate full automation is achieved. The invention also provides a kit which can realize recycling of the marker and is long in luminescence time, high in intensity and easy to determine, so that the detection sensitivity is improved, the linear range is widened, and the dilution error of a high-value specimen is reduced.
