Problems solved by technology

However, long-term use of these agents is associated with an increased risk of cardiovascular events, raising concerns about the high risk-benefit ratio for recommending these agents for CRC chemoprevention
Other agents including folic acid, calcium, vitamin D, and antioxidants have also been studied for chemoprevention, but their efficacy remains to be determined

[0080] Example 1: Streptococcus thermophilus and its inhibitory effect on CRC

[0081] illustrate

[0082] The inventors of the present application have identified Streptococcus thermophilus and the molecules it secretes as new prophylactic or therapeutic agents for the prevention or treatment of CRC. More specifically, the inventors of the present application demonstrated that co-cultivation of Streptococcus thermophilus with colon cells can inhibit cancer cell proliferation both in vitro and in vivo. Compounds secreted by bacteria can have the same effects in vitro and in vivo. In addition, secreted molecules can induce programmed cell death and cause cell cycle arrest in G0 / G1 phase. Administration of this specific probiotic prevented the formation of CRC in animals. Therefore, Streptococcus thermophilus and the molecules it secretes can be used in the prevention and treatment of CRC.

Abstract

The present application provides novel methods of treating and preventing colorectal cancer in a subject by administering a composition comprising beta-galactosidase, a bacterial culture (e.g., a Streptococcus thermophilus culture) capable of producing beta-galactosidase, or a culture component comprising beta-galactosidase. The invention further provides a set product for the method. In addition, the present application provides a composition for treating or preventing colon cancer.

Description

[0001] related application [0002] This application claims priority to U.S. Provisional Patent Application No. 62 / 799,162, filed January 31, 2019, the contents of which are hereby incorporated by reference in their entirety for all purposes. [0003] Background of the invention [0004] Colorectal cancer (CRC) is the third most common cancer and the fourth leading cause of cancer death worldwide, with an estimated one million new cases and >500,000 deaths per year. According to data published by the Hong Kong Hospital Authority Cancer Registry, CRC accounted for 16.4% of all new cancer cases and 14.3% of all cancer deaths in Hong Kong in 2012. [0005] Several intrinsic factors (eg, age, male sex, diabetes, obesity, and inflammatory bowel disease) and extrinsic factors (eg, smoking, insufficient fiber intake, high intake of alcohol, red meat, and high-fat diet) are associated with increased risk of CRC related. The epidemiological characteristics of CRC are in a dynamic s...
