Abstract

The embodiment of the specification relates to a method and a kit for detecting Escherichia coli. In the detection scheme, an upstream primer 16F19 and a downstream primer 16R1514 are designed and synthesized based on a homologous sequence of 16SrRNA in Escherichia coli, an upstream primer LF20 and a downstream primer LR1039 are designed and synthesized based on a homologous sequence of LacI geneof a lactose operon in Escherichia coli K-12 MG1655, and a dual PCR reaction system for amplification and identification is constructed based on the combination of the primers. The use of the dual PCRdetection not only ensures specificity, but also is simple to operate, and avoid the influence of complex factors; and the use of 16SrRNA as a control indicator ensures that Escherichia coli is not missed, and false positives can be controlled because the LacI gene is a unique gene of Escherichia coli. Therefore, the detection sensitivity and specificity are improved, and at the same time, the detection is simple, fast, low-cost and easy to promote.

Description

technical field [0001] The embodiment of this specification relates to the technical field of biological detection, in particular to a method and a kit for detecting Escherichia coli. Background technique [0002] Escherichia coli (Escherichia coli, E.coli), also known as Escherichia coli, is one of the indicators of fecal contamination. Among the three indicators of fecal contamination, coliforms, heat-resistant coliforms and Escherichia coli, the detection of Escherichia coli is the most significant. Therefore, Escherichia coli is a frequently tested indicator in sample testing. The number of Escherichia coli in the water body reflects the situation that the water body is polluted by human and animal feces. The limit standard for Escherichia coli in drinking water in my country is not to be detected (GB5479-2006). There are three methods for the detection of Escherichia coli in drinking water issued by the "National Standard of the People's Republic of China" (hereinaft...
