Embodiment approach

[0021] In addition, the Metrn1 protein can also be artificially prepared, such as prepared according to conventional genetic engineering techniques. Any suitable Metrnl protein is suitable for use in the present invention. The Metrn1 protein includes a full-length Metrn1 protein or a biologically active fragment thereof. As a specific embodiment, the Metrn1 protein may be a human Metrn1 protein with a size of about 30 kDa, and the amino acid sequence is shown in NP_001004431.1.

[0022] The amino acid sequence of Metrn1 protein formed by substitution, deletion or addition of one or more amino acid residues is also included in the present invention. The Metrn1 protein or its biologically active fragment includes a part of conservative amino acid substitution sequence, and the amino acid substitution sequence does not affect its activity or retains its partial activity. Appropriate substitution of amino acids is well known in the art and can be readily performed without alteri...

[0031] Example D-Galactose-induced dementia model preparation and Metrn1 protein involved in the regulation of brain cognitive function

[0032] 1. Experimental method

[0033] 1. Breeding of genetically engineered experimental animals. Metrnl systemic knockout mice were obtained by mating and breeding using the Cre-LoxP conditional gene knockout system (Diabetes 2015; 64:4011-4022). The mating and breeding process requires three kinds of mice, the source of which is: Metrnl loxP / loxP Exclusively owned by us (Diabetes 2015; 64:4011-4022), Ella-Cre mice were purchased from Shanghai Nanfang Model Biotechnology Development Co., Ltd., and C57 mice were purchased from Shanghai Xipro-Bikai Experimental Animal Co., Ltd. After mating and breeding, Metrn1 systemic knockout mice (knock out, KO) and wild type normal control mice (wild type, WT) can be obtained at the same time.

Abstract

The invention relates to an application of Metrnl protein or a Metrnl gene to the respect of preventing and treating cognitive disorder. According to the application disclosed by the invention, a Metrnl systemic knockout mouse and a corresponding wild type contrast mouse are adopted, a D-galactose induced dementia model is prepared, learning and memory functions are detected through a Morris watermaze experiment, the result displays that compared with the wild type contrast mouse, the learning and memory functions of the Metrnl knockout mouse are reduced, protein expression of brain-derived neurotrophic factors and the like in a hippocampus is reduced, and it is indicated that the Metrnl can be used for preventing and treating cognitive dysfunction type diseases. A new method is providedfor preventing and treating cognitive disorder; and the Metrnl is a human body endogenous substance and possibly has small side effects on human bodies, so that the safety of the Metrnl as a potentialmedicine is very high.

Description

technical field [0001] The invention relates to the technical field of medicine, specifically, the application of Metrn1 protein or gene in the prevention and treatment of cognitive impairment. Background technique [0002] Cognition is the intelligent processing process for the body to recognize and acquire knowledge, involving learning, memory, language, thinking, spirit, emotion and other behaviors. Cognitive impairment refers to the abnormality of the brain's advanced intelligence processing process related to the above-mentioned learning, memory and thinking judgment, which causes learning and memory impairment. Cognition is a normal function of the brain, and any factors that cause abnormalities in brain function and structure can lead to cognitive impairment. As the world continues to accelerate towards the aging society, diseases such as cognitive impairment and senile dementia (Alzheimer's disease) have become important risk factors that threaten the health of huma...
