Problems solved by technology

The incidence of leg and foot disease in domestic large-scale pig farms is about 5%-35% (Yu Yongjun, 2015), and the mortality rate of sows in affected pig herds may exceed 5% (Zhang Jiayi, 2010). The resulting sows were eliminated or died early, which brought huge economic losses to the pig farm

Embodiment 1

[0024] Example 1: Pig Genotyping Detection

[0025] (1) Using the kit method to extract ear tissue DNA from bone density-related populations

[0026] The kit in this example uses the TIANamp Genomic DNA Kit Blood / Cell / Tissue Genomic DNA Extraction Spin Column Kit, produced by Tiangen Biochemical Technology (Beijing) Co., Ltd. The specific steps are:

[0027] 1) The ear-like tissues of bone density-related populations (the breeds are Large White pigs and Landrace pigs, and the implementation of the present invention is not limited to the above breeds, which are provided by Guangxi Yangxiang Co., Ltd.), are cut into paste with ophthalmic scissors, Add 200ulGA (included with the above kit), shake and mix well; then put it into a 56°C water bath for overnight digestion.

[0028] 2) Add 200ul buffer GB (included with the above kit) to the digested tissue sample, mix thoroughly by inversion, and place at 70°C for 10 minutes, the solution should become clear, and centrifuge briefly...

Embodiment 2

[0037]Example 2: Application of molecular marker typing method in association analysis of bone density traits of pig limbs and hooves

[0038] (1) Correlation analysis between ASGA0099279 molecular marker typing results and bone density traits of limbs and hooves

[0039] The test pigs used for the correlation detection and analysis of genotype and bone density traits of limbs and hooves come from purebred sow groups bred by Guangxi Yangxiang Co., Ltd., including Large White pigs and Landrace pigs (conventional breeds, but not limited to the above breeds) . The DNA used for genotyping is extracted from ear samples of purebred Large White pigs and Landrace pigs ("Purebred Large White pigs and Landrace pigs" referred to in the text of the instructions and tables are referred to as "pigs" for short). Using the method based on the single-marker association regression model, using the gender of the individual as a fixed effect, the GWAS analysis was performed using the GenABEL sof...

Abstract

The invention belongs to the field of animal molecular marker screening, and particularly relates to an SNP molecular marker of an FAM131C gene associated with bone density of a limb and hoof of a sow. An FAM131C gene segment of a gene upstream and downstream 100bp sequence with an accession number of ASGA0099279 is obtained by a genotyping technology and referring to an Ensembl database, and thesequence is as shown in SEQ ID NO:1. By conducting typing on the FAM131C gene using a gene chip technology, associated markers of the limb and hoof of the sow are obtained by screening, the sequence is as shown in SEQ ID NO:1, and the R of the 101 basic group of the sequence is C or T, which leads to the polymorphism of a nucleotide sequence. The gene can be taken as the molecular marker usedfor the detection associated to the sow bone density, and when the 101 nucleotide on SEQ ID NO:1 is C, the limb and hoof of the sow have higher bone density. The invention provides a new SNP marker for marking assisting selection of sow bone density.

Description

technical field [0001] The invention belongs to the technical field of animal molecular marker screening, in particular to a SNP molecular marker of the FAM131C gene related to the bone density of sow limbs and hooves. The molecular marker of the invention can be used for detection of bone density traits of pig limbs and hooves. Background technique [0002] my country is a big pork consumer, and the hog industry is an important part of our national economy. In order to improve the production level of our country's hogs and meet the growing domestic demand for lean meat, a large number of lean meats such as Duroc, Landrace, and Large White are introduced from abroad every year. Meat type pig. In my country's intensive breeding environment, these pigs are prone to limb and foot disease, lameness and even paralysis, resulting in early elimination of breeding pigs. Leg disease in sows can delay estrus or affect the symptoms of estrus, prolong the time of empty pregnancy, sows ...
