Problems solved by technology

There are a large amount of genetic information data of Dendrobium in the NCBI database, and there have been many reports on the variation characteristics of ITS gene sequences and their application in phylogenetic relationship and species identification, but the comparison of previous research samples on Dendrobium less, can not achieve good repeatability, making the data and conclusions not comprehensive, not perfect, and even contradictory
In the Dendrobium genus, some common species are easy to distinguish, such as Dendrobium drumsticks, Dendrobium officinale, Dendrobium swollen and Dendrobium bark, etc., but many species are very similar in shape and difficult to identify

Embodiment 1

[0030] Embodiment 1 The construction of Dendrobium plant barcode

[0031] 1. Collection of Dendrobium plant samples

[0032] In the present invention, 109 samples of 12 Dendrobium species were collected in the early stage, and each species has 8-10 samples, so that it has better repeatability, so that the identification of Dendrobium plants by DNA barcodes is more real and effective. The collected samples of Dendrobium species were all confirmed by plant morphological identification, see Table 1 for details.

[0033] 2. Extraction of sample DNA

[0034] 0.5g of each sample was extracted according to the improved CTAB method (Doyle J. DNA protocols for plants-CTABtotal DNA isolation. In: Hewitt GM, Johnston A, eds. Molecular techniques intaxonomy. Berlin: Springer, 1991, 283-293.) Genomic DNA (see Chapter 2). After the extracted DNA is purified by RNase, the optical density values at 260nm and 280nm are measured with a UV spectrophotometer. When the DNA purity reaches OD26...

Abstract

The invention relates to a method for identifying dendrobium plant and belongs to the technical field of molecular marker. The method comprises the following steps: extracting total DNA from a sample;using an ITS5F / 4R primer for amplifying and then using CodonCode Aligner for checking and splicing a sequencing peak figure of the amplifying product; removing low-mass sequence and primer area; using ClustalXu2.0 for comparing multiple sequences and then removing the redundant sequences at the two ends; using Mega6.0 software for calculating the K-2-P distance of the treated sequence; analyzingand comparing intra-species inter-species genetic distance of the sequence and establishing a phylogenetic tree, thereby distinguishing the dendrobium species by clustering the ITS sequences of different dendrobiums. The method provided by the invention is capable of solving the technical problem in the prior art that the traditional methods, such as phenotypic traits or physical and chemical analysis, are difficult to identify the dendrobium plant and is capable of quickly, conveniently and accurately identifying the dendrobium plant.

Description

technical field [0001] The invention belongs to the technical field of molecular markers, and in particular relates to a method for identifying plants of the genus Dendrobium. Background technique [0002] Dendrobium is one of the largest genus of Orchidaceae (Orchidaceae), and there are 74 species and 2 varieties of Dendrobium in my country. Dendrobium dendrobium is sweet in nature and slightly cold; it belongs to the stomach and kidney channels; it has the effects of benefiting the stomach, promoting body fluid, nourishing yin and clearing away heat. Dendrobium nobile, Dendrobium officinale officinale and Dendrobium versicolor (Dendrobium fringe); the 2010 edition stipulates that Dendrobium is Dendrobium nobile and Dendrobium drumstick chrysotoxum Or the fresh or dried stems of cultivated products of Dendrobium fringe and its similar species of plants of the same genus, and Dendrobium candidum is listed as a separate species; the 2015 edition stipulates that dendrobiu...
