Abstract

The invention belongs to the technical field of body immunity, and discloses a measuring method for inducing death of an antigen specificity CD4<+>T cell. The measuring method uses an IEC-source Gal-1 and simultaneously activates a T cell receptor to induce; the measuring method comprises the following steps: preparing an OVA specificity CD4<+>T cell; separating a non-specificity CD3<+>CD4<+>CD25<->T cell from a small intestine of a normal B6 mouse; preparing Gal-1 defective IEC, detecting the effect through immunoblotting; culturing the cell, putting the IEC on a top chamber and putting the CD4<+>T cell on a bottom chamber when culturing the cell through a permeable insulating chamber; and analyzing FACS data through Flowjo software. The measuring method shows that: after the IEC absorbs an antigen, the Gal-1 is likely to be produced, the processed antigen information is transferred to the T cell, the T cell receptor is activated, and the activation induced cell death is induced. The Gal-1 produced by the IEC is likely to strengthen AICD (Activation Induced Cell Death).

Description

technical field [0001] The invention belongs to the technical field of body immunity, and in particular relates to a detection method for inducing apoptosis of antigen-specific CD4+ T cells. Background technique [0002] Immune tolerance refers to the specific immune non-response state produced by the body's immune system after accepting the action of a certain antigen. Individuals who have developed immune tolerance to a specific antigen do not develop an immune response or cannot develop an immune response that can be detected by conventional methods after being stimulated by the antigen again. Therefore, the normal immune tolerance mechanism is very important to maintain the health of the body. The mechanism of immune tolerance is still not very clear. Although the international and domestic research on the factors that impair immune tolerance has made rapid progress in recent years, the restoration of the established immune tolerance damage and the recovery of the resu...
