Problems solved by technology

[0003] However, although a large number of lamb eggs can be obtained by hormone induction, the quality of these eggs is uneven, resulting in the development ability of lamb embryos in vitro is much lower than that of adult sheep embryos

Embodiment 1

[0042] Embodiment 1, the method for improving the development ability of lamb embryo

[0043] 1. Configuration of culture medium for promoting egg maturation

[0044] 1. Collection of follicular fluid from adult sheep

[0045] Follicular fluid can be collected from hormone-induced or non-hormone-induced adult sheep.

[0046] 1) Collection of hormone-induced follicular fluid from adult sheep

[0047] The hormone induction program adopts the routine superovulation procedure of sheep, and the hormones used include follicle-stimulating hormone (FSH) or pregnant horse gonadotropin (PMSG).

[0048] Hormone-inducing regimen: inject follicle-stimulating hormone FSH (Ningbo Sansheng) to adult ewes 4 times in 2 consecutive days or 6 times in 3 days, with an interval of 12 hours each time, a total of 480IU FSH (Ningbo Sansheng). 12 hours after the last injection, it can be taken from a living body or extracted from an isolated ovary after slaughter. Specifically, a 10G needle disposab...

Abstract

The invention discloses a method for improving the developmental capacity of lamb in-vitro embryos and provides a method for accelerating in-vitro maturation of lamb ova. The method comprises the following steps: carrying out in-vitro maturation culture of in-vitro lamb cumulus-oocytes complexes in an ovum maturation accelerating culture solution added with adult lamb follicular fluid, and achieving the in-vitro maturation of the lamb ova. Through the method, the ovum maturation accelerating culture solution added with adult lamb follicular fluid is used for culturing the lamb oocytes; after the ova are mature, the ova can produce in-vitro embryos through in-vitro fertilization; the developmental capacity of the lamb embryos can be obviously improved; the method is simple and feasible and is convenient in material sources.

Description

technical field [0001] The invention belongs to the field of embryo engineering, in particular to a method for improving the development ability of lamb in vitro embryos. Background technique [0002] Lamb embryo in vitro production technology (Juvenile in vitro Embryo Transfer, JIVET) is a comprehensive technology that integrates superovulation, live oocyte collection, egg in vitro maturation, in vitro fertilization, embryo in vitro culture and embryo transfer. Presexual lambs are used as egg donors to produce in vitro embryos. Studies have shown that the number of follicle development in young lambs (4-8 weeks old) is more than 10 times that of adult sheep using the same hormone treatment regimen. Thus, young lambs are an effective egg donor for the production of in vitro embryos. Moreover, using young lambs as egg donors can also advance the breeding age of sheep by more than one year. Therefore, JIVET technology is a rapid breeding technology with great application va...
