Embodiment 1

[0019] Observe the three-dimensional structure of the trachea inside the barbel of the bare-shouldered star.

[0020] Preparation of fixative: use 0.2M phosphate buffer to dilute 5% glutaraldehyde solution p Adjust the H value to 7.2~7.4; use 0.2M phosphate buffer to dilute the 1% osmium tetroxide p Adjust the H value to 7.2~7.4.

[0021] Pretreatment: Pre-cool the long-shouldered longhorn beetle at 4°C for 4 hours, and then quickly cut off its mandibular barbels with a blade.

[0022] Fixative solution treatment: quickly put the excised mandible into the prepared glutaraldehyde solution to soak and fix for 48 hours, then wash with 0.2M phosphate buffer solution for 2 hours, and then soak and fix with the prepared osmium tetroxide solution for 1 hour after cleaning , and then washed with distilled water for 2 hours, and finally blotted the surface liquid with filter paper, embedded in paraffin, and then observed on the machine.

Embodiment 2

[0025] Observe the three-dimensional structure of the trachea inside the lower lip palp of the bare-shouldered star.

[0026] Preparation of fixative: use 0.2M phosphate buffer to dilute 5% glutaraldehyde solution p Adjust the H value to 7.2~7.4; use 0.2M phosphate buffer to dilute the 1% osmium tetroxide p Adjust the H value to 7.2~7.4.

[0027] Pre-treatment: Pre-cool the long-shouldered beetle at 4°C for 4 hours, and then cut off its lower palps quickly with a blade.

[0028] Fixative solution treatment: use the prepared glutaraldehyde solution to continuously perfuse and fix the freshly excised lower lip for 48 hours (you can continuously perfuse fresh glutaraldehyde solution, or circulate the quantitative glutaraldehyde solution through a peristaltic pump); Then wash with 0.2M phosphate buffer solution for 2 hours, soak and fix with the prepared osmium tetroxide solution for 1 hour after cleaning, then wash with distilled water for 2 hours, and finally use filter paper ...

Abstract

The invention provides a method for observing the three-dimensional structure of the trachea of an insect by micro CT (Computed Tomography). The method comprises the following steps: a, sample treatment: performing tissue fixation on a respiratory organ of a target insect by using a fixing solution at the temperature of 4 DEG C; b, micro CT observation: observing a treated sample by micro CT. The method has the advantages of observing the three-dimensional characteristic of the respiratory organ, rebuilding and observing the structure of any plane, along with no tissue damage, high speed, high efficiency and high repeatability. By adopting the method, the basic structure of the insect can be known comprehensively from outside to inside and from a plane to three dimension, and an important basis and a technical measure are provided for comprehensive observation of the internal respiratory system structure of the insect.

Description

technical field [0001] The invention relates to an observation method of insect respiratory organs, in particular to a method for observing the three-dimensional structure of insect trachea by using micro-CT. Background technique [0002] The respiratory system of insects is an important gas exchange organ of insects. The research on the structure of insect respiratory organs is very important both from the perspective of biological evolution and from the perspective of insect control. At present, there are two methods commonly used to study insect respiratory organs, paraffin section method and transmission electron microscope method. [0003] Paraffin section method, also known as cell section method, is the most commonly used method for observing tissue cells. The morphology and structure of cells can be clearly identified by using paraffin section method. In particular, this method can be used for targeted staining, which can only be used for the neurons that need to be ...
