The compound biological substrate is combined with separable laminated structure and blocky structure, wherein the laminated structure comprises a top layer, a fiber layer I, a water storage layer I, a fiber layer II, a water storage layer II, a bottom layer and a waterproof layer in turn from top to bottom. According to the compound biological substrate for greening roof and the preparation method thereof provided by the invention, the compound biological substrate has a certain structure according to the difference in the rooting depth of the plant in different growth stages, so that the purposes of preserving moisture and fertility and boosting the continuous healthy growth of the plant can be achieved.
