Problems solved by technology

Synthetic organic germanium is hydroxyethyl germanium sesquioxide, namely Ge-132, spirogermanium, furan germanium derivatives and other anti-virus, anti-inflammatory, anti-cancer organic germanium, but taking too much can easily cause calcium deficiency; natural organic germanium Germanium is extracted from natural plants and can be eaten directly. Its disadvantages are high cost and very limited output; biological organic germanium enriches germanium compounds in organisms, such as yeast, bacteria and large fungi.

Embodiment 1

[0056] Embodiment 1: strain screening

[0057] Candida utilis (Candida utilis) (purchased from China Industrial Microorganism Culture Collection and Management Center, strain number: CICC 1314) was inoculated into germanium-containing medium, and the strains were respectively treated under appropriate conditions. The species were subcultured (subcultured for 12 to 18 generations) for domestication and their enrichment of germanium was detected. Finally, a strain with high ability to tolerate germanium, transform germanium and enrich germanium is obtained, that is, the domesticated Candida utilis.

[0058] The formula of the germanium-containing medium: molasses 50-150g / L, urea 3-10g / L, germanium dioxide 500mg / L-2000mg / L.

[0059] The cultivation conditions: temperature 25°C-35°C.

Embodiment 2

[0060] Embodiment 2: the preparation method of germanium-rich yeast

[0061] The production process is as follows: slant strain cultivation→first-level liquid seed cultivation→secondary liquid seed cultivation→fermentation tank seed cultivation→fermentation tank cultivation→separation→drying→packaging.

[0062] (1) Slant strain culture: the Candida utilis obtained after the acclimation of Example 1 is transferred to the potato sucrose medium (sucrose 30g / L, peptone 10g / L, yeast powder 10g / L, germanium dioxide 50mg / L) L, agar 20g / L, pH value 4.8~6.5), activated on a solid plate, cultured at a constant temperature of 25~35°C for 45 hours, picked a single colony slant and preserved it for later use.

[0063] (2) First-level liquid seed culture: Pick 1-4 rings of yeast strains on the slope and inoculate them in 100-500mL potato sucrose liquid medium (sucrose 30g / L, peptone 10g / L, yeast powder 10g / L, germanium dioxide 50mg / L, pH value 4.8-6.5) in the Erlenmeyer flask, at 25-35 ℃ c...

[0070] Embodiment 3: the preparation method of germanium-rich yeast

[0071] The production process is as follows: slant strain cultivation→first-level liquid seed cultivation→secondary liquid seed cultivation→fermentation tank seed cultivation→fermentation tank cultivation→separation→drying→packaging.

[0072] (1) Slant strain culture: the Candida utilis obtained after domestication in Example 1 is transferred to the potato sucrose medium (sucrose 20g / L, peptone 4g / L, yeast powder 5g / L, germanium dioxide 100mg / L) L, agar 20g / L, pH value 4.8~6.5), activated on a solid plate, cultured at a constant temperature of 28°C for 48 hours, picked a single colony slope and preserved it for later use.

[0073] (2) First-level liquid seed culture: Pick 1-4 rings of yeast strains on the slope and inoculate them in 100-500mL potato sucrose liquid medium (sucrose 20g / L, peptone 4g / L, yeast powder 5g / L, germanium dioxide 100mg / L, pH value 4.8 ~ 6.5) in the Erlenmeyer flask, at 28 ℃ constant ...

Abstract

The invention discloses germanium-enrichment yeast and a preparation method application thereof. The preparation method disclosed by the invention comprises the steps: firstly, subculturing and domesticating candida utilis in a germanium-contained culture medium to obtain domesticated candida utilis; then culturing the domesticated candida utilis in a saccharomycete fermentation medium to obtain the germanium-enrichment yeast. According to the preparation method disclosed by the invention, the germanium-enrichment yeast with high biological activity and low toxicity is finally obtained by utilizing germanium enrichment of the candida utilis, utilizing capacity of low-cost raw materials and utilizing molasses and urea as fermentation raw materials. By means of the preparation method, the germanium-enrichment yeast product with higher organic germanium purity and yield can be finally obtained. The germanium content of per kilogram of germanium-enrichment yeast product is 837.1 to 1206.4mg, and the organic germanium content is 93.3% to 98.6%. By means of the preparation method, inorganic germanium can be converted into organic germanium, so that organic germanium source is widened, and large-scale application of the germanium in the field of food, fodder and the like is improved.

Description

technical field [0001] The invention belongs to the field of food and feed additives, and relates to a germanium-rich yeast and its preparation method and application. Background technique [0002] Germanium, as a trace element, was discovered by German chemist Winkler in 1886. In 1929 Dutoit discovered germanium in animals. Trace amounts of germanium are present in many enzymes (eg, guaninase, cytochrome oxidase), cerebral cortex, and gray matter components in animals. After the antibacterial activity of trialkylgermanium acetate was discovered in 1962, the biological effects of organic germanium attracted people's attention. At present, some compounds of germanium have been proved to have various biological activities. [0003] Biological functions of germanium: [0004] (1) Participating in immune regulation Aso found through mouse experiments that germanium can significantly increase the content of interferon in mouse serum. According to reports by Tang Zhaoxin et a...
