Embodiment 1

[0032] Example 1 Preparation of Bacillus circulans Y3 bacterial agent

[0033] 1. Isolation and purification of Bacillus circulans Y3

[0034] A strain Y3 that can effectively control sunflower sclerotinia and rust was isolated and purified from the farmland soil of Gongzhuling, Jilin Province, my country.

[0035] According to the mycological characteristics of the strain Y3 and the 16S rDNA sequencing result (SEQ ID NO: 1), the strain Y3 was determined to be Bacillus circulans.

[0036] 2. Cultivation of Bacillus circulans Y3

[0037] (1) Composition of medium (kg / t fermentation liquid)

[0038] Soluble starch 10kg, soybean cake powder 10kg, corn flour 5kg, glucose 1kg, sucrose 10kg, yeast powder 5kg, (NH 4 ) 2 SO 4 3kg,K 2 HPO 4 0.2kg, NaCl 2.5kg, MgSO 4 ·7H 2 O 0.1kg, CaCO 3 0.5kg, FeSO 4 0.001kg, boric acid (1%) 1L, pH7.0, prepared with water.

[0039] (2) Fermentation and cultivation process

[0040] Bacillus circulans Y3 was inoculated on potato sucrose (P...

Embodiment 2

[0045] Example 2 Growth-promoting effect of Bacillus circulans Y3 bacterial agent on potted corn

[0046] 1. The inoculation test of Y3 bacterial agent was carried out under greenhouse conditions, and the soil came from the farmland soil of Tangjialing. Corn was planted under pot conditions, and the biofertilizer bacterial agents prepared by the two strains were inoculated respectively. The treatments were CK, ACCC 10094 and Y3, respectively, and the strain ACCC 10094 was a potassium-solubilizing bacterium, which was used as a control strain. The results showed that both ACCC 10094 and Y3 strains had the effect of promoting crop growth, and both could increase the biomass of maize (Table 1). The fresh weight biomass of corn treated with strain ACCC10094 and Y3 reached 16.2g / pot and 18.4g / pot respectively, which were 28.6% and 46.0% higher than the control; the dry weight biomass of strain ACCC10094 and Y3 reached 1.504g / pot and 1.8626g respectively / pot, compared with the c...

Embodiment 3

[0050] The growth-promoting effect of embodiment 3 Y3 microbial agent to potted rapeseed

[0051] The potting test soil comes from Tangjialing farmland soil. Rapeseed is planted under potting conditions and inoculated with bio-fertilizer bacteria prepared by ACCC10094 and Y3 strains respectively. The fresh weight of rapeseed treated with ACCC10094 and Y3 bacterial agents reached 5.475g / pot and 7.66g / pot respectively, which were 73.3% and 142.4% higher than the control; the dry weight of rapeseed with ACCC10094 and Y3 bacterial agents was 1.036 g / pot and 1.276g / pot, respectively increased by 38.9% and 71.0% compared with the control. The bacterial agent prepared by Y3 strain significantly increased the biomass of rapeseed.

[0052] Table 2 The effect of Y3 bacterial agent on the biomass of potted rapeseed

Abstract

The invention provides a Bacillus cereus Y3 and an application thereof in the control of crop soil-borne diseases. The preservation number of the Bacillus cereus Y3 is CGMCC NO. 13511, the Bacillus cereus Y3 is processed to prepare microbial agents, the microbial agents have a substantial control effect on sunflower soil-borne diseases sclerotinia and rust and have an obvious growth promotion effect on corn, tomato, rape, sunflower and other crops, the yield increase and disease control effects of the microbial agents are not inhibited by a chemical fertilizer, and the microbial agents are used to continuously increase the yield under normal chemical fertilizer use conditions. The sunflower sclerotinia induced rotten disk disease control effect of the microbial agents reaches 95% or above, the rust control effect reaches 100%, and the sunflower yield increases by 20-50%. A result of pot experiments shows that the microbial agents promote the growth of the crops, and improve the biomass amount by 15-70%. The microbial agents are of great significance to control the soil-borne soil diseases of farmland, continuously increase the yield of grains and protect the ecological environment of the farmland.

Description

technical field [0001] The invention relates to the technical fields of microbiology and plant disease biocontrol, in particular to a strain of Bacillus circulans Y3 and its application in preventing and controlling crop soil-borne diseases. Background technique [0002] With the improvement of crop yield and the development of facility agriculture, the problems of soil-borne diseases and continuous cropping obstacles have become increasingly prominent, and soil pathogenic microorganisms have continued to increase, causing crop yield reduction or even failure. Soil-borne diseases and continuous cropping obstacles have become bottlenecks in the development of safe agricultural production. Continuous cropping for 3 years will reduce yield by 10% to 20%, and crop yield will decrease by 40% to 50% in continuous cropping for 4 to 5 years. Soil-borne diseases of protected vegetables, potatoes, garlic, sunflower, rapeseed, cotton, soybean, rice, wheat, and sugar beet occur frequen...
