Embodiment 1

[0025] The design scheme of the ploidy mutagenesis experiment of amisulalin acting on the germinated thorn grape seeds is shown in Table 1: the experiment was carried out by dipping method. Taking clear water dipping as a contrast, set 3 repetitions, 12 treatments in total; each germination box is filled with 500ml of each treatment solution, soaking 100 thorn grape seeds that have germinated to the dewy stage with roughly the same effect of germination acceleration, and the soaking time is respectively 12h, 24h, 36h. The content of DMF as the organic solvent of amisulfameline in the treatment solution is 0.04%-0.08%.

Abstract

The invention discloses a ploidy mutagenesis method of dinitramine-dipped brier grape seeds. The method comprises the following steps: (1) dissolving dinitramine in dimethylformamide, and diluting via water until the mass concentration of dinitramine is 0.0025%-0.0035% to obtain a dinitramine solution; (2) dipping the brier grape seeds in a white bud emerging stage in the dinitramine solution for 12-36 hours to complete ploidy mutagenesis of the brier grape seeds. Through the ploidy mutagenesis method of dinitramine-dipped brier grape seeds, the brier grape seeds are successively induced to obtain brier grape tetraploids according to a dipping method; the dipping solution is dinitramine; the optimal concentration is about 0.003%; the mutagenesis rate reaches 60% or higher.
