Embodiment 1

[0018] Mix 3mL of n-butyl titanate with 10mL of ethanol to obtain a uniform solution A; dissolve 0.06g of o-phenanthroline in 10mL of ethanol to obtain a uniform solution B; slowly add solution B to A under constant stirring solution, a homogeneous solution was obtained. The obtained solution was added into a hydrothermal reaction kettle, and reacted at 180° C. for 24 hours. Centrifuge the obtained solid, wash it with deionized water and ethanol alternately for more than three times, dry it, and grind it. Mix 50mg of the powder obtained by grinding with 20mL of 1uM CYP119 enzyme solution, stir at 10°C for 12h, centrifuge, and wash to obtain the product.

Embodiment 2

[0020] Mix 6mL n-butyl titanate solution with 20mL ethanol to obtain a uniform solution A; dissolve 0.12g o-phenanthroline in 20mL ethanol to obtain a uniform solution B; slowly add solution B to A under constant stirring solution, a homogeneous solution was obtained. The obtained solution was added into a hydrothermal reaction kettle, and reacted at 180° C. for 24 hours. Centrifuge the obtained solid, wash it with deionized water and ethanol alternately for more than three times, dry it, and grind it. Mix 100mg of powder obtained by grinding with 40mL of 1uM CYP119 enzyme solution, stir at 10°C for 12h, centrifuge, and wash to obtain the product.

Abstract

The invention relates to a method for degrading methyl orange by means of photo-catalytic oxidation by the aid of CYP119 enzymes loaded with TiO2-phenanthroline. The methyl orange which is an organic pollutant can be effectively degraded by materials obtained by the aid of the method. The method includes uniformly mixing tetrabutyl titanate solution and phenanthroline solution with each other and carrying out reaction on the tetrabutyl titanate solution and the phenanthroline solution in a hydrothermal kettle; separating samples after reaction is completed, cleaning, drying and grinding the samples and then mixing and stirring the samples and the CYP119 enzymes with one another to obtain products. The method has the advantages that TiO2 modified by the phenanthroline which is an organic matter is used as a carrier for the CYP119 enzymes, and the materials are efficient in photo-catalytic oxidation performance.

Description

technical field [0001] The invention relates to the technical field of photocatalysis and biocatalysis, in particular to a method using TiO 2 - A method for the photocatalytic oxidation degradation of methyl orange loaded with o-phenanthroline and CYP119 enzyme. Background technique [0002] The CYP119 enzyme is a cytochrome P450 enzyme with exceptional stability and tolerance. Its thermal denaturation temperature is as high as 90°C, and it can withstand high pressure above 200MPa and extreme pH conditions. It is an excellent biocatalyst and an ideal target for protein engineering. It has extremely important applications in the fields of biopharmaceuticals and biosynthesis. value. Recent studies have shown that it has a special photoactivation mechanism, which provides a basis for the construction of a photocatalytic system based on CYP119 enzymes, making it possible to replace complex and expensive auxiliary enzyme systems with simple and cheap photocatalytic systems, whi...
