Problems solved by technology

[0002] At present, although there have been reports on the application of DNA barcoding technology for plant identification, and certain normative documents have been formed (Gao Lianming et al., "Technical Specifications for Plant DNA Barcoding Research", Journal of Plant Taxonomy and Resources, 2012, 34 (6): 592 ~ 606), however, if DNA barcoding technology is to be applied to the identification of mahogany, it will face prominent difficulties different from conventional plant classification:
[0003] Question 1: Data sources for establishing a DNA barcode sequence database
[0004] Question 2: The selection of the target gene of plant DNA barcoding has always been a key factor affecting the reliability of this technology for species identification
[0005] Question 3: Algorithms for DNA barcode database construction and discriminant criteria for unknown samples
Even for any one of the methods, the parameters used in the analysis are different, and the results may be different

Embodiment Construction

[0023] A DNA barcode technology for identifying redwood and its products, using the strategy of merging the experimental data of redwood samples and the database data of target plants to build a high-capacity database: Sequencing the DNA barcode sequences of redwood samples, and merging the sequencing sequences into the Genbank database has been published The same sequence of redwood species was constructed to construct a DNA barcode gene database with a wide range of data sources, and the matK+rbcL gene was used as the target DNA barcode gene for redwood identification. The genetic distance method based on -parameter probability and the phylogenetic tree method based on cluster analysis establish the identification database algorithm and judgment rules to comprehensively improve the accuracy and credibility of identification.

[0024] refer to figure 1 As shown, the DNA of 18 redwood samples was used as a template, and rbcL-specific primers were used for PCR amplification. Ex...

Abstract

The invention discloses a DNA bar code technology for identifying rosewood and rosewood products and an application method thereof. The technology is as below: constructing a high capacity database through a strategy by merging database of rosewood samples experiment data and a target plant; sequencing a DNA bar code sequence of the rosewood samples; merging the sequencing sequences with homologous sequences published in the Genbank database to construct a rosewood DNA bar code gene database with wide range of data sources; and establishing an algorithm and decision rules for the identification database by using matK+rbcL genes as the target DNA bar code gene for rosewood identification, and using a search tool method of a local alignment algorithm based on frequency, a genetic distance method based on Kimura-2-parameter probability and a phylogenetic tree method based on clustering analysis. The DNA bar code technology for identifying rosewood and rosewood products and the application method thereof establish an accurate and quick rosewood DNA bar code identification technology.

Description

technical field [0001] The invention relates to a DNA barcode technology for identifying mahogany and its products and its application method. Background technique [0002] At present, although there have been reports on the application of DNA barcoding technology for plant identification, and certain normative documents have been formed (Gao Lianming et al., "Technical Specifications for Plant DNA Barcoding Research", Journal of Plant Taxonomy and Resources, 2012, 34 (6): 592 ~ 606), however, if DNA barcoding technology is to be applied to the identification of mahogany, it will face prominent difficulties different from conventional plant classification: [0003] Question 1: The source of data for establishing a DNA barcode sequence database. Since mahogany is a common category named from the perspective of consumption, there is no clear species relationship between various species, and there are great differences in the source and rarity of different mahogany species, s...
