Embodiment Construction

[0025] The present invention will be described in detail below in conjunction with the examples.

[0026] 1) Establishment of Peeled catfish family

[0027] The parents of wild yellow catfish were collected from Honghu Lake, Dongting Lake, Zhongxiangnan Lake and Changhu Lake for crossbreeding. From the offspring F1 population, 18 males and 18 females were randomly selected for artificial induced breeding. The ratio of males and females was 1:1, and 18 A full sibling family. The fin ray tissues of the parents of each family were clipped and placed in absolute ethanol, and the family information was recorded and stored at -20°C for later use. 18 families were placed in 18 plastic water tanks for separate breeding. After the fry hatched for 30 days, about 10 fish were randomly selected from each family, fixed with absolute ethanol, and used as samples for family identification.

Abstract

The invention discloses a pelteobagrus fulvidraco microsatellite family identification method which comprises steps as follows: pelteobagrus fulvidraco parent and offspring genomic DNA extraction, polymorphic microsatellite marker screening and primer synthesis, microsatellite locus genotyping and family identification and the like. Six pairs of microsatellite primers are adopted and have nucleotide sequences shown from SEQ ID NO. 1 to SEQ ID NO.12 respectively. Fluorescently-labeled microsatellite markers are firstly utilized on pelteobagrus fulvidraco, a paternity test platform is established, the accurate rate of identification of a pelteobagrus fulvidraco microsatellite family is up to 98.9%, different families of pelteobagrus fulvidraco can be rapidly and effectively identified, and evidence is provided for breeding and breeding matching of pelteobagrus fulvidraco. The pelteobagrus fulvidraco microsatellite family identification method selects more microsatellite locus alleles with high polymorphism, can be used for population genetics evaluation, family tree authentication and paternity test of pelteobagrus fulvidraco and further can be used for molecular marker auxiliary family management and molecular marker auxiliary parent selection.

Description

technical field [0001] The invention relates to a molecular marker auxiliary technology for fish genetic breeding, in particular to a method for identifying a family of yellow catfish by using fluorescent microsatellite markers. Background technique [0002] Pelter catfish is an important economic fish in rivers and lakes in my country. It has strong adaptability to the ecological environment and is widely distributed in fresh water bodies in my country. Because of its tender meat, rich nutrition, high meat content, and no intermuscular spines except spines, it is very popular among consumers. In recent years, overfishing has become more and more serious because the commercial yellow catfish mainly relies on natural fishing. In addition, lakes and rivers are increasingly polluted by pesticides and industrial waste water, etc., and the nature of the yellow catfish has been damaged to a certain extent. It is imperative to protect the resources of the yellow catfish and relate...
