Embodiment 1

[0029] The detection method provided in this embodiment comprises the following steps:

[0030] 1. Preparation of Prussian blue-labeled molecular antigen:

[0031] Small molecular compounds with active groups such as amino groups or carboxyl groups are coupled to chitosan molecules through amidation reaction (EDC / NHS reaction), diazotization method, glutaraldehyde method, etc., and by adjusting the pH value, the Chitosan labeled with small molecular compounds is positively charged, and then ferrous sulfate and potassium ferricyanide are added to its dispersion, and negatively charged Prussian blue is synthesized on its surface, and the labeling of small molecule antigens by Prussian blue is realized through electrostatic interaction .

[0032] 2. Preparation of magnetic antibody:

[0033] Add ferrous chloride and ferric chloride at a ratio of 1:2, dissolve them in water and heat to 70°C, add sodium hydroxide solution with 8 times the molar number of ferrous chloride, mix qui...

Abstract

The invention relates to the field of electrochemical immunological techniques, and particularly relates to a method for detecting a small molecule compound based on a prussian blue bionic marker. The method comprises the following steps: using an analogue of a prussian blue micro-particle marker to-be detected object with catalytic activity like that of catalase as a marker antigen; immobilizing an antibody with a magnetic ferroferric oxide particle, dispersing the magnetic ferroferric oxide particle in a reaction liquid; and forming an oxidation current signal in a competitive immune reaction and electrochemical detection system and further realizing quantitative measurement on the to-be-detected object. According to the method, a small molecule antigen is molecularly marked by adopting prussian blue instead of a bio-enzyme and a competence immune reaction is performed by combining with the magnetic particle, thus the high-sensitivity quantitative detection on the small molecule compound can be realized through an electric signal generated by a catalytic oxidation reaction of the electrochemical detection marker antigen to hydrogen peroxide and paradioxybenzene because the size of the electric signal is in linear dependence with the concentration of a to-be-detected small molecule.

Description

technical field [0001] The invention relates to the technical field of electrochemical immunity, and relates to a method for detecting small molecule compounds based on Prussian blue bionic markers. Background technique [0002] In the current field of biomedical detection, high-sensitivity quantitative immunoassays for small molecules such as environmental pollutants, pesticides, hormones, and antibiotics have become one of the focuses of researchers. In order to achieve high-sensitivity detection of small or trace amounts of small molecular compounds, various methods and systems based on biospecific reactions, such as antigen-antibody technology and protein ligand technology, have been established. Among them, the typical one is the competitive immunoassay technology, which uses the competition between the labeled antigen and the analyte to be tested against the antibody, and the concentration of the analyte to be tested contained in the system can be deduced by detecting ...
