Embodiment

[0032] Example: The method for separating and purifying chlorogenic acid and 3,5-dicaffeoylquinic acid from honeysuckle, the steps are:

[0033] (1) Preparation of the crude extract of honeysuckle: Take the crushed honeysuckle, heat and reflux extraction with 70% ethanol (adjust the pH to about 4 with hydrochloric acid), filter, and concentrate the ethanol extract under reduced pressure to obtain the crude extract.

[0034] (2) Extraction: Dissolve the above crude extract in hot water at about 70°C, add hydrochloric acid to adjust the pH to about 2, then extract with ethyl acetate, and the ethyl acetate extract is concentrated under reduced pressure to obtain total phenols of honeysuckle acid extract.

[0035] (3) Separation and purification of macroporous adsorption resin: Put the total phenolic acid extract of honeysuckle on a macroporous adsorption resin column, first elute with water, and then use 20%, 40%, 60%, 80%, 95% ethanol-water solution for Gradient elution in th...

Abstract

The invention relates to a method for separating and purifying chlorogenic acid and 3,5-dicaffeoylquinic acid from a honeysuckle flower. According to the method, the honeysuckle flower is taken as a raw material and the method comprises the following steps: (1) preparing a honeysuckle flower crude extract; (2) extracting; (3) separating and purifying through macroporous adsorption resin; (4) separating and purifying through a semi-preparation type high efficiency liquid chromatography, namely separating and purifying by adopting the semi-preparation type high efficiency liquid chromatography to obtain two high-purity components by taking methanol-water as a mobile phase, wherein the two high-purity components are respectively identified as the chlorogenic acid and the 3,5-dicaffeoylquinic acid. The process disclosed by the invention is environment-friendly in process without severe harm on the environment and is low in integrated cost.

Description

technical field [0001] The invention belongs to the field of chemical industry, in particular to a method for separating and purifying chlorogenic acid and 3,5-dicaffeoylquinic acid from honeysuckle. Background technique [0002] Honeysuckle is the honeysuckle plant Lonicera ( Lonicera japonica Thumb.) dry buds or flowers with early blooming, cold in nature, sweet in taste, belonging to the lung, heart, and stomach meridian, has the functions of clearing away heat and detoxifying, cooling and dispelling wind-heat, and is used to treat acute tonsillitis, pharyngitis, mastitis, Dysentery, pneumonia, influenza, hepatitis, appendicitis and other diseases. Honeysuckle contains phenolic acids, flavonoids, saponins, and volatile oils, among which phenolic acids (including chlorogenic acid, caffeic acid, and isochlorogenic acid) are the main active ingredients. Isochlorogenic acid is a mixture of 7 isomers with very similar structures, among which 3,5-dicaffeoylquinic acid is the...
