Embodiment 1

[0026] This embodiment includes the following steps:

[0027] (1) Take 100 grams of mixed phytosterol, 51.1 grams of β-sitosterol, and 200 grams of n-pentanol, mix and heat to 65°C, stir evenly, heat filter to remove impurities, and obtain the filtrate; cool the filtrate to 40°C, stir at constant temperature for 2 hours, and keep warm Filter to get 62.5g of filter cake, which is the crude product of β-sitosterol;

[0028] In the mixed phytosterols, the total sterol mass content is 96.3%, wherein β-sitosterol is 50.4%, and stigmasterol is 16.2%; the β-sitosterol is refined sitosterol with a purity of 91.0%;

[0029] (2) Continue to cool down the filtrate obtained in step (1) to 20° C., stir at constant temperature for 3 hours, and heat-filter to obtain the filtrate and filter cake (30.5 g). The filter cake is crude mixed sterols;

[0030] (3) Heat and dissolve the crude mixed sterols obtained in step (2) with 120g of n-amyl alcohol, prepare a saturated solution of stigmasterol...

Embodiment 2

[0035] This embodiment includes the following steps:

[0036] (1) Take 100 grams of mixed phytosterol, 51.1 g of β-sitosterol, and 180 g of n-pentanol, mix and heat to 65°C, stir evenly, and heat filter to remove impurities to obtain the filtrate; the filtrate is cooled to 40°C, stirred at constant temperature for 3 hours, and then filtered , to obtain filter cake 65.9g, filter cake is β-sitosterol crude product;

[0037] The mixed phytosterols have a total sterol mass content of 95.1%, including 60.4% β-sitosterol and 10.7% stigmasterol; the β-sitosterol is refined sitosterol with a purity of 92.3%;

[0038] (2) Continue to cool down the filtrate obtained in step (1) to 20° C., stir at constant temperature for 2 hours, and heat-filter to obtain 24.8 g of filter cake, which is crude mixed sterols;

[0039] (3) Heat and dissolve the crude mixed sterols obtained in step (2) with 100g of n-amyl alcohol, prepare a saturated solution of stigmasterol at 50°C, add 3g of water (20°C)...

Abstract

A method for segmented purification of beta-sitosterol and stigmasterol by using n-amyl alcohol comprises the following steps: (1) mixing and dissolving blended phytosterol and n-amyl alcohol at the temperature of 60-70 DEG C, then adding beta-sitosterol to prepare a beta-sitosterol saturated solution at the temperature, thermally filtering, cooling a filter liquor, stirring at a constant temperature, preserving heat and filtering to obtain a crude product of beta-sitosterol; (2) continuing to cooling the filter liquor, stirring at the constant temperature, preserving heat and filtering to obtain a crude product of blended sterol; (3) heating and dissolving the crude product of blended sterol by using the n-amyl alcohol to prepare a stigmasterol saturated solution, cooling, stirring at the constant temperature, preserving heat and filtering to obtain a crude product of stigmasterol; (4) heating and concentrating the filter liquors in the step (2) and the step (3), cooling, stirring at the constant temperature, preserving heat and filtering to obtain the crude product of beta-sitosterol; (5) refining the crude product of stigmasterol by using the n-amyl alcohol; (6) refining the crude product of beta-sitosterol by using the n-amyl alcohol. The method provided by the invention is simple in steps, a single solvent is easy to recover and does not cause pollution, and the product purity and the yield are high.

Description

technical field [0001] The invention relates to a method for segmentally purifying β-sitosterol and stigmasterol by using n-pentanol. Background technique [0002] Phytosterol is a natural active substance present in plants, mainly including β-sitosterol, stigmasterol and campesterol. They have the same chiral structure, and the only difference in structure is the side chain. β-sitosterol, also known as sitosterol, has the effects of lowering cholesterol, relieving cough, anticancer, and anti-inflammation; it can be used as a drug for regulating hypercholesterolemia, alleviating atherosclerosis, and preventing and treating prostate diseases. It has obvious curative effect on breast cancer and can also be used as an anti-inflammatory drug and a drug against chronic acetic acid gastric ulcer. Stigmasterol has the function of significantly lowering human serum cholesterol, preventing cerebral hemorrhage and arteriosclerosis. It is used for blood lipid regulation and anti-ather...
