Problems solved by technology

However, the current problems in the tissue culture of honeysuckle limit the formation and development of the honeysuckle industry
The main problems are: ①difficult sterilization of explants; ②short subculture period (1-2 months) and no one does in vitro preservation; ③low survival rate of test tube transplantation and high cost of test tube seedling cultivation; ④test tubes for tissue culture Seedlings are rarely used in production on a large scale. Only Li Hong et al. (2007) report on industrialized production and cultivation management technology of honeysuckle tissue culture. However, the overall level of industrialized seedling cultivation is not high, and the key production link technology is weak, resulting The low reproduction coefficient greatly limits the popularization and application of famous and precious varieties
So far, there are no reports on the rapid propagation and in vitro preservation methods of honeysuckle seedlings in the prior art

Embodiment 1

[0015] 1. Establishment of sterile clones

[0016] Material scientific name: Lonicera japonica Thnnb.

[0017] Sterilization of explant materials is an important link in tissue culture, because honeysuckle stems are hollow and covered with sparse pilose and sparse glandular hairs, it is difficult to sterilize at the initial stage of tissue culture. It is required to completely kill the microorganisms on the surface of the outer material, but also to damage the outer material tissue and surface cells as little as possible. The ideal effect has been achieved through the research on the sterilization process. The specific methods are as follows:

[0018] 1. Select high-yield and high-quality Lonicera japonica Thnnb. live plants (compact plant type, plants with about 30 flowering buds in each leaf axil) in the field or in the field, move them into pots and place them indoors for maintenance, capping first (remove The top stem tip) is beneficial to the growth of axillary buds, wat...

Abstract

The invention provides a method for rapid propagation and in-vitro conservation of honeysuckle seedlings. The method comprises the following steps: establishment of a sterile and clone system, rapid propagation, rooting culture, fake planting and transplantation of test-tube seedlings and in-vitro conservation. The honeysuckle has high health-care function, ornamental function, medical function and ecological function, is one of special famous and precious medicines in China and enjoys good reputation of penicillin in traditional Chinese medicines. The invention provides a solution for the problems in the tissue culture of the honeysuckle and an optimal culture technology. The method provided by the invention has the advantages that the propagation speed is 1: (4-6), the survival rate of transplantation is more than 90%, 99% rooting effect is obtained by matched use of paclobutrazol (PP333) and indolebutyric acid (IBA), and the heredity of the obtained test-tube seedling generated by sprouts is stable; the sterile and clone system of the honeysuckle established by the method has no pollution in the culture process, the growth of explants on the culture medium is restored rapidly, and technical support is provided for large-scale and industrial production of the high-quality honeysuckle seedlings.

Description

[0001] Field: [0002] The invention belongs to the field of biotechnology, and in particular relates to a method for rapid propagation and in vitro preservation of honeysuckle seedlings. Background technique: [0003] Honeysuckle is a species with high health care, ornamental, medicinal and ecological functions. It is one of the unique and precious Chinese medicinal materials in my country. It has the reputation of "penicillin" in traditional Chinese medicine. Especially the role played in the prevention and treatment of SARS in 2003 has attracted people's attention, and it has become a key development object among Lonicera resource plants. Modern pharmacological studies have shown that honeysuckle has broad-spectrum antibacterial, antiviral, antitumor, immune-enhancing and antipyretic, anti-inflammatory, choleretic and liver-protecting, blood-lipid-lowering, anti-fertility, hemostatic and anti-ulcer and other biological activities. Production and sales are booming in the ma...
