However, the R30 strain can only grow normally in an acidic environment of pH 3 at most. Therefore, when the pH of the bioleaching system is less than 3, the bacteria will be inhibited and its stability will be affected.

Embodiment Construction

[0020] Examples of the present invention are described below.

[0021] 1. Isolation and expanded culture of strain D13

[0022] Prepare PDA medium, ingredients: 200g potatoes, 20g sucrose or glucose, 1000mL water, natural pH, sterilized at 121°C for 30min. The samples used to isolate acid-resistant heterotrophic bacteria were taken from the acidified sludge (pH value around 2.0) after the previously completed municipal sludge bioleaching treatment. Use an inoculation loop to take a loop of the acidified sludge solution, streak and separate it on a PDA plate, and culture the above plate inoculated with the sludge suspension upside down at 28°C for 3-4 days until colonies grow. Separate a representative single colony with good growth on the PDA plate for smear microscopy. If there is impurity, then pick the colony and repeatedly streak until the pure bacteria of the target strain are obtained, and transfer the slant to save it for later use. Liquid culture conditions: Take the...

Abstract

The invention relates to an acid resistant heterotrophic bacterium D13 applied in bioleaching of municipal sludge, which belongs to the field of environmental engineering. A unicell of the bacterium is oval, has a diameter of 4.5 to 5.5 mu m, has one end for budding, generates no ascospores and has capsule. A bacterium colony of the bacterium is in a form of mucilage and white and has wrinkles onthe surface and regular edges. The bacterium can grow normally in an extreme acidic environment that the pH value is 2. According to the morphology, physio-biochemical characteristics and molecular biology identification of the bacterium, the heterotrophic bacterium belongs to pichia spartinae and is assigned an accession number of CGMCC NO.3035. In an bioleaching environment for sludge, the bacterium can rapidly use water-soluble small molecule organic matters or simple organic acid in the sludge as a carbon source and energy and enables the content of CO2 in bioleaching solution to be increased substantially; therefore, bioleaching efficiency of sludge is improved, the removal rate of heavy metals in the sludge reaches more than 90%, and the dehydration property of the sludge is improved by more than ten times.

Description

1. Technical field [0001] The invention relates to an acid-resistant heterotrophic bacterium D13 applied to urban sludge bioleaching treatment, which is a technology for sludge bioleaching treatment by utilizing the interaction between heterotrophic bacteria and autotrophic bacteria, and belongs to the technical field of environmental engineering . 2. Technical Background [0002] my country produces a large amount of various types of urban sludge and industrial sludge that need to be disposed of urgently every year. At present, only municipal sludge produces about 7 million tons (in terms of dry matter) every year, and the amount of industrial sludge is also very large. The operating cost of sludge treatment and disposal accounts for 30%-50% in sewage treatment plants, and these sludge There is no good way to deal with it in our country at present. On the one hand, it is difficult to concentrate and dehydrate sludge, and the water content of dewatered sludge is very high ...
