The samples used to isolate acid-resistant heterotrophic bacteria were taken from the acidified sludge (pH value around 2.0) after the previously completed municipal sludge bioleaching treatment. Use an inoculation loop to take a loop of the acidified sludge solution, streak and separate it on a PDA plate, and culture the above plate inoculated with the sludge suspension upside down at 28°C for 3-4 days until colonies grow. Separate a representative single colony with good growth on the PDA plate for smear microscopy. If there is impurity, then pick the colony and repeatedly streak until the pure bacteria of the target strain are obtained, and transfer the slant to save it for later use.
