specific Embodiment

[0020] Taking different explants as an example, the best induction effect was achieved by using different media and different culture conditions. In the following, different explants are used as objects to elaborate respectively:

[0021] Induction of embryonic callus:

[0022] Cultivation period: every September, after the seeds are formed;

[0023] Pre-cultivation treatment: use a combination of low temperature (4°C) treatment and gibberellin treatment, and use conventional disinfection methods (2% NaClO for 30 minutes, 70% alcohol for 30 seconds, rinse with sterile water for 3-5 times) after disinfection vaccination;

[0024] The best medium: MS+NAA0.1mg / L+6-BA1.0mg / L;

[0025] The callus induction rate reached 100%.

[0026] Stem segment callus induction:

[0027] Material collection period: take young stems, in late March and early April in spring, or young stems formed by germination of seeds;

[0028] Pre-cultivation treatment: use conventional sterilization metho...

Abstract

The invention discloses a method for inducing callus of peony petals, applying an MS culture medium that contains 6-BA cytokinin, NAA plant growth regulator, 2, 4-D weedicide and the like to induce callus. Specific culture medium scheme and hormone combination are supplied for explants of peony petals. The optimal material drawn period of the callus is described in the invention. The inductivity of the peony callus is more than 80% and a preferable induction effect is achieved; and the inductivity of the petal callus is higher than 50%.

Description

[0001] This application is a divisional application of: [0002] Application number: 200910083744.3; Invention name: method for inducing peony callus; application date: 2009.5.11. technical field [0003] The invention relates to a method for culturing callus, in particular to a method for inducing callus from peony petals. Background technique [0004] Peony is one of the top ten famous flowers in China, and it is also the national flower of China. It has the reputation of "the king of flowers". Tissue culture of tree peony mainly uses buds to induce plants and explants to induce plants through callus, and the callus induction of tree peony is an important stage for plant regeneration, and a higher callus induction rate is more important for later callus. How to obtain a higher callus induction rate is closely related to the type of medium, the type and concentration of hormones, and the culture conditions. [0005] Callus culture is the most common form of culture. Excep...
