Abstract

The invention relates to a method for detecting alga and colored dissolved organic matters by adopting a flow cytometry. The method comprises the following steps of: (1) alga detection, namely irradiating particles to be detected by a laser beam keeping an angle of 90 degrees with the particles, detecting a forward scattering light (FWC), a side scattering light (SWC) and multi-color fluorescent light of the particles by a detection system, and automatically analyzing by utilizing software, so as to obtain related information such as the fluorescent signal strength of an alga cell, the size of the alga cell and the quantity of the alga cells in unit volume; and (2) detection on colored dissolved organic matters, namely filtering the detected alga particles, irradiating filtrate by a laser beam with the wavelength of 355nm, collecting a light absorption signal and a fluorescent signal of the filtrate in a special wavelength range by the detection system, and carrying out analysis on the colored dissolved organic matters by an intelligent instrument. The method provided by the invention is simple and rapid, the alga and the colored dissolved organic matters (CDOM) in water can be simultaneously continuously detected online, and early warning detection on eutrophication of a water body can be realized.
