Embodiment Construction

[0026] see first figure 1 , is a device for applying oscillatory constant fluid shear force to adherent cells in the present invention. The device is composed of a microliter syringe 3 , a connecting tube 4 , a small fluid chamber 5 , a silicone sealing ring, an oscillating fluid loading device 2 and a single-chip microcomputer 1 . The microliter syringe 3 is connected to the liquid inlet and outlet 12 at one end of the fluid small chamber 5 through the connecting tube 4; the other connecting tube 4 is connected to the liquid inlet and outlet 12 at the other end of the fluid small chamber 5, and inserted into the liquid storage bottle . The microliter syringe 3 is fixed in the mounting groove 6 of the microliter syringe group in the oscillating fluid loading device 2, that is, the whole device is formed. The reciprocating movement of the plunger of the microliter syringe 3 forms an oscillating fluid shear force applied to the adherent cells.

Abstract

The invention discloses a device for applying an oscillatory constant fluid shearing force to anchorage-dependent cells, which comprises a microliter injector, connecting tubes, a small fluid chamber, an oscillatory fluid loading device and a single chip, wherein the microliter injector is connected with a fluid inlet and a fluid outlet at one end of the small fluid chamber through one connecting tube; the other connecting tube is connected with the fluid inlet and the fluid outlet at the other end of the small fluid chamber and is inserted into a fluid storage bottle; and when the oscillatory fluid loading device works, the injection volume and injection speed of the injector can be uniformly and accurately controlled, so that the flow rate and oscillation frequency of reciprocating fluid in the oscillatory fluid loading device are controlled. The device of the invention has a simple structure, is convenient to operate, can form a sealed environment and also can prevent the pollution of bacteria and adjust the oscillation frequency and the magnitude of the shearing force in experimental research.

Description

technical field [0001] The invention belongs to the field of biomechanics and relates to a cell mechanical loading device, in particular to a device for applying oscillatory constant fluid shear force to adherent cells cultured in vitro. Background technique [0002] Fluid shear stress is a kind of stress that generally exists in blood flow and bone tissue in the living body. For example, for bone tissue in poroelastic media, mechanical loading will cause deformation of the bone matrix and the fluid flow channels existing in the bone voids, resulting in fluid flow in the voids, thereby generating fluid shear stress. This fluid shear stress is oscillatory. [0003] In the article entitled Shear stress induced stimulation of mammalian cell metabolism published by JA Frangos et al. in 1988, a parallel plate flow chamber was first proposed, which was widely used in cell mechanics experiments. It is mainly used to study the adhesion between cells and cells, cells and substrate ...
